Measuring DNA modifications with the comet assay: a compendium of protocols

Andrew Collins et al.

Summary: The comet assay is a versatile method for detecting various types of DNA damage in individual eukaryotic cells. It has been widely used in research, industry, and regulatory agencies, and has been adopted as an in vivo test for genotoxicity. The present document provides consensus protocols for applying the comet assay to different cell types, species, and types of DNA damage.

DNA damage response signaling: A common link between cancer and cardiovascular diseases

Sepideh Nikfarjam et al.

Summary: DNA damage response (DDR) signaling is crucial for maintaining genomic and proteomic homeostasis and dysregulation in DDR pathways is associated with various pathophysiological states, including cancer and cardiovascular diseases (CVDs). This review comprehensively discusses different DDR pathways, the cross talk among DDR mechanisms, the role of DDR in cancer and CVDs, the similarities and differences between DDR in cancer and CVDs, the role of DDR in the pathophysiology of CVDs, interventional strategies for targeting genomic instability in CVDs, and future perspectives.

Adherence to Mediterranean Diet in Croatia: Lessons Learned Today for a Brighter Tomorrow

Marko Geric et al.

Summary: Non-communicable diseases (NCD) and lifestyle, particularly diet, are closely related. The study evaluates adherence to the Mediterranean diet and its association with demographic and anthropometric data. The results show that women, residents of coastal areas, older individuals, those with higher education and income, and lower BMI are associated with higher adherence scores. Additionally, a dietary plan following Mediterranean diet principles is found to be cost-effective. These findings provide important insights for encouraging lifestyle changes.

DNA damage in circulating leukocytes measured with the comet assay may predict the risk of death

Stefano Bonassi et al.

Summary: The comet assay, a common method for measuring strand breaks and other DNA lesions, was used in a study to assess the impact of DNA damage on overall mortality and disease risk. The results showed that individuals with high levels of DNA damage had a significantly higher overall mortality and risk of circulatory system diseases. These findings support the use of the comet assay in preventative strategies for non-communicable diseases.

The hCOMET project: International database comparison of results with the comet assay in human biomonitoring. Baseline frequency of DNA damage and effect of main confounders

Mirta Mili et al.

Summary: The alkaline comet assay is widely used for evaluating DNA damage in human population, but faces challenges due to large inter-individual and inter-laboratory variation. The hCOMET project aims to establish reference values and compare different sources of assay variability, with the analysis of data from multiple countries, laboratories, and years leading to significant findings.
