Enantioselective Deaminative Alkylation of Amino Acid Derivatives with Unactivated Olefins

Summary: Here, we report the first Ni-catalyzed enantioselective deaminative alkylation of amino acid and peptide derivatives with unactivated olefins. A new sterically encumbered bis(oxazoline) ligand backbone was discovered, offering a de novo technology for accessing enantioenriched sp(3)-sp(3) linkages via sp(3) C-N functionalization. This protocol has a broad scope and generality, even applicable for late-stage functionalization. Additionally, an enantioselective deaminative remote hydroalkylation reaction of unactivated internal olefins is achievable, providing an entry point for forging enantioenriched sp(3)-sp(3) centers at remote sp(3) C-H sites.

Synthesis of Flavanone and Quinazolinone Derivatives from the Ruthenium-Catalyzed Deaminative Coupling Reaction of 2′-Hydroxyaryl Ketones and 2-Aminobenzamides with Simple Amines

Summary: A highly effective catalyst, the cationic Ru-H complex, was found to facilitate the deaminative coupling reaction of 2'-hydroxyaryl ketones with simple amines to form flavanone products. Similarly, the reaction of 2-aminobenzamides with branched amines yielded quinazolinone products. This catalytic method efficiently installs useful core structures without the need for reactive reagents.

Three-Component Ruthenium-Catalyzed meta-C-H Alkylation of Phenol Derivatives

Summary: In this study, we successfully demonstrated the multicomponent reactions of phenol derivatives via a six-membered cycloruthenated intermediate for the first time. This strategy showed good substrate suitability and functional group tolerance, providing a potential synthetic drug approach. Mechanistic studies suggested the involvement of a radical in the process. Additionally, the meta alkylated phenol was obtained by further removal of the directing group.

Deaminative Cyclization of Tertiary Amines for the Synthesis of 2-Arylquinoline Derivatives with a Nonsubstituted Vinylene Fragment

Summary: By using triethylamine as a vinylene source, a convenient protocol for the regioselective synthesis of beta,gamma-nonsubstituted 2-arylquinolines from aldehydes and arylamines has been developed. The deaminative cyclization can also be extended to long-chain tertiary alkylamines, allowing diverse alkyl groups to be simultaneously introduced into the pyridine rings. This process demonstrates a new conversion pathway for simultaneous dual C(sp(3))-H bond functionalization of tertiary amines, in which the transient acyclic enamines generated in situ undergo the Povarov reaction.

Dual Nickel/Photoredox-Catalyzed Deaminative Cross-Coupling of Sterically Hindered Primary Amines

Scope and Mechanism of the Redox-Active 1,2-Benzoquinone Enabled Ruthenium-Catalyzed Deaminative α-Alkylation of Ketones with Amines

Summary: Nitrogen heterocycles are prevalent in drugs and nonplanar heterocycles with stereogenic centers are of significant interest in medicinal chemistry, chemical biology, and synthetic methods development. A Rh-catalyzed C-H bond alkenylation/electrocyclization cascade has been developed to access highly functionalized nitrogen heterocycles with multiple stereogenic centers, providing versatile intermediates for elaboration with high regioselectivity and stereoselectivity. Mechanistic experiments and DFT calculations support the reactions, which can lead to the synthesis of complex polycyclic products, including bioactive molecules such as the opioid antagonist (-)-naltrexone.

Synthesis of Cyclopentenones through Rhodium-Catalyzed C-H Annulation of Acrylic Acids with Formaldehyde and Malonates

Summary: An efficient rhodium-catalyzed protocol for the synthesis of cyclopentenones based on a three-component reaction of acrylic acids, formaldehyde, and malonates was reported. The treatment of a variety of alkyl halides with a Na2CO3/MeOH solution could smoothly achieve 5-alkylation of the cyclopentenones. Excess formaldehyde and malonate led to a multicomponent reaction that afforded multisubstituted cyclopentenones through a Michael addition.

Cobalt(III)-Catalyzed Diastereo- and Enantioselective Three-Component C-H Functionalization

A three-component C-H functionalization catalyzed by an earth-abundant Co(III) complex with high enantioselectivity has been described, providing a rapid access to substituted beta-hydroxyketones. The outlined reactivity of (CpCo)-Co-x(III) catalysis shows a higher and exploitable propensity for selective additions across carbonyls compared to Rh(III) chemistry.
