Carboxylic Acids as Building Blocks in Catalytic Asymmetric Reactions

Nilanjana Majumdar

Summary: Catalytic asymmetric transformation is a powerful strategy for synthesizing diverse nonracemic molecular entities. Carboxylic acids have been explored as building blocks in this area, and significant advancements have been made in the last three decades. They can act as both C- and O-nucleophiles, as well as electrophiles, in asymmetric reactions under catalytic conditions.

Asymmetric Catalytic Reactions of Donor-Acceptor Cyclopropanes

Yong Xia et al.

Summary: The donor-acceptor (D-A) cyclopropanes, due to the synergistic push-pull effect of vicinal electron-donating and electron-withdrawing groups, are recognized as powerful building blocks for generating polyfunctional reactive intermediates. Enantioselective reactions of D-A cyclopropanes provide an efficient approach to enantioenriched acyclic and cyclic compounds. Chiral Lewis/Bronsted acids, transition metals, and organocatalysts have been designed for such transformations, including ring-openings, annulations, and rearrangements.

Summary: The study established a reaction under the cooperative bimetallic catalysis of iridium and copper catalysts, which yielded indole C3-allylation products with moderate yields (up to 78%) and good enantioselectivities (up to 97:3 er). This reaction not only represents the first catalytic asymmetric ring-opening reaction of vinylcyclopropanes with C3-unsubstituted indoles, but also provides an atom-economic and straightforward method for the synthesis of C3-allylic indoles with high regio- and enantioselectivity.
