Chiral Photocatalyst Structures in Asymmetric Photochemical Synthesis

Matthew J. Genzink et al.

Summary: Asymmetric photocatalysis is a major research topic in contemporary synthetic organic chemistry. However, the discovery of general strategies for highly enantioselective photochemical reactions has been a recent development, leading to a somewhat limited variety of stereocontrolled photoreactions. This review summarizes the chiral catalyst structures that have been studied for solution-phase asymmetric photochemistry, providing insight into the logic required for highly effective stereocontrolled photocatalysis.

Visible Light-Induced Regio- and Enantiodifferentiating [2+2] Photocycloaddition of 1,4-Naphthoquinones Mediated by Oppositely Coordinating 1,3,2-Oxazaborolidine Chiral Lewis Acid

Nao Shimizu et al.

Summary: A range of asymmetric photochemical transformations using visible light have recently become considerably attractive. Chiral Lewis acid association to enones for [2 + 2] and ortho photocycloadditions and oxadi-pi-methane rearrangements have shown to be very promising in achieving effective enantiodifferentiation. In this study, we investigated the regio- and enantiodifferentiation of [2 + 2] photocycloaddition reactions of naphthoquinone derivatives mediated by chiral oxazaborolidines. We found that the excited-state decomplexation is a key factor in achieving high regio- and enantioselectivities in the photocycloaddition reactions.

Robust Analytical Methods for Monitoring the Formation of a Chiral Oxazaborolidine Catalyst

Sloan Ayers et al.

Summary: Having reliable methods to monitor catalyst activation processes is crucial for ensuring the reproducibility of catalytic reactions. For asymmetric Diels-Alder reactions, chiral oxazaborolidine or oxazaborolidinium catalysts are powerful reagents that promote these reactions with high yield and selectivity. By investigating the mechanism, several modern analytical methods are compared to quantitate the formation of oxazaborolidine catalyst from amino alcohol and boroxine, leading to useful monitoring methods for this important transformation.

Enantioselective Construction of Vicinal Angular Quaternary Stereocenters Enabled by Strained Cyclobutenones

Peng Yan et al.

Summary: In this study, we report a new method for enantioselective Diels-Alder reaction catalyzed by a chiral catalyst COBI, which enables the asymmetric reaction of 2,3-disubstituted cyclobutenones with various dienes. This method successfully synthesizes bicyclo[4.2.0]octane derivatives with two contiguous angular quaternary centers, demonstrating intriguing structural patterns.

London Dispersion Interactions Rather than Steric Hindrance Determine the Enantioselectivity of the Corey-Bakshi-Shibata Reduction

Christian Eschmann et al.

Summary: The well-known Corey-Bakshi-Shibata (CBS) reduction is a powerful method for the asymmetric synthesis of alcohols, with London dispersion (LD) interactions playing a crucial role in determining enantiodiscrimination. By modifying CBS catalysts with dispersion energy donors (DEDs), it was possible to improve catalyst design for challenging CBS reductions.

Catalytic Asymmetric Darzens-Type Epoxidation of Diazoesters: Highly Enantioselective Synthesis of Trisubstituted Epoxides

Dong Guk Nam et al.

Summary: The highly enantioselective Darzens-type epoxidation of diazoesters with glyoxal derivatives was achieved using a chiral boron-Lewis acid catalyst and a chiral oxazaborolidinium ion catalyst, leading to the asymmetric synthesis of trisubstituted alpha,beta-epoxy esters with high yield (up to 99%) and excellent enantio- and diastereoselectivity (up to >99% ee and >20:1 dr, respectively). The synthetic potential of the method was demonstrated by the conversion of the products to various compounds such as epoxy gamma-butyrolactone, tertiary beta-hydroxy ketone, and epoxy diester.
