Metallaphotoredox: The Merger of Photoredox and Transition Metal Catalysis

Amy Y. Chan et al.

Summary: The merger of photoredox catalysis with transition metal catalysis, known as metallaphotoredox catalysis, has become an important part of synthetic methodology. It combines the bond formation capability of transition metal catalysis with the utility of photoinduced electron and energy transfer processes. Photocatalytic substrate activation allows the involvement of simple starting materials in metal-mediated bond-forming processes, while electron or energy transfer with organometallic intermediates provides a complementary activation mode to traditional catalytic platforms.

Merging Halogen-Atom Transfer (XAT) and Copper Catalysis for the Modular Suzuki-Miyaura-Type Cross-Coupling of Alkyl Iodides and Organoborons

Zhenhua Zhang et al.

Summary: We report a mechanistically distinct approach for Suzuki-Miyaura-type cross-coupling reactions between alkyl iodides and aryl organoborons. This method utilizes the halogen-atom-transfer ability of alpha-aminoalkyl radicals to activate secondary alkyl iodides, which are then coupled with various aryl, vinyl, alkynyl, benzyl, and allyl boronate species.

Cobalt-Catalyzed C(sp2)-C(sp3) Suzuki-Miyaura Cross-Coupling Enabled by Well-Defined Precatalysts with L,X-Type Ligands

L. Reginald Mills et al.

Summary: In this study, efficient precatalysts for the C(sp(2))-C(sp(3)) Suzuki-Miyaura cross-coupling reaction between alkyl bromides and neopentylglycol (hetero)arylboronic esters were generated in situ using Cobalt(II) halides in combination with phenoxyimine (FI) ligands. The protocol allowed for efficient C-C bond formation with various nucleophiles and electrophiles. The results also suggested that alkyl radicals play an important role in the catalytic reaction.

A general N-alkylation platform via copper metallaphotoredox and silyl radical activation of alkyl halides

Nathan W. Dow et al.

Summary: A catalytic union of various functional groups with electronically and sterically diverse alkyl bromides has been achieved through a visible-light-induced metalla-photoredox platform, bypassing traditional high barriers. This regio- and chemoselective protocol is compatible with a wide range of medicinally relevant N-nucleophiles and structurally diverse alkyl bromides. Preliminary mechanistic experiments validate the dual catalytic, open-shell nature of this platform, enabling reactivity previously unattainable in traditional halide-based N-alkylation systems.

Summary: The study demonstrates an alternative amination strategy using alkyl iodides as radical precursors and copper catalysis to assemble sp3 C-N bonds at room temperature. This method provides S(N)2-like programmability and has been shown to be useful in the late-stage functionalization of pharmaceuticals for the preparation of valuable N-alkylated drug analogues.
