GPCR-mediated β-arrestin activation deconvoluted with single-molecule precision

Wesley B. Asher et al.

Summary: Beta-arrestins are strongly autoinhibited in their basal state but can be activated by phosphorylated receptors and receptor agonism, releasing the beta-arrestin tail and allowing it to assume different conformations. The rate and efficiency of beta-arrestin activation depend on the phosphorylation pattern of the receptor and agonist-promoted receptor activation.

Structure of the vasopressin hormone-V2 receptor-β-arrestin1 ternary complex

Julien Bous et al.

Summary: Recent structural studies have provided insights into the molecular mechanisms of GPCR-arrestin coupling, but it is unclear whether this process is conserved among different GPCRs. In this study, the cryo-electron microscopy active structure of the wild-type arginine-vasopressin V2 receptor (V2R) in complex with beta-arrestin1 was determined, revealing a unique arrangement of beta-arrestin1 and an interface involving all receptor intracellular loops. These findings suggest a significant structural variability among GPCR-arrestin signaling complexes.

Biochemical insights into structure and function of arrestins

Yasmin Aydin et al.

Summary: Arrestins are multifunctional cytosolic adaptors that bind to active and phosphorylated GPCRs via a versatile interface, stopping G protein signaling and triggering other pathways. Recent studies have provided a wealth of information about the key determinants and dynamic insights into the arr-GPCR interaction, but questions about the functional consequences of structural details and interaction dynamics remain open for further investigation. Techniques such as mutagenesis studies, cross-linking studies, conformational probes, and sensors have been used to explore arr structure, function, and dynamics, offering valuable insights into the process of arr binding and the outcomes of different binding modes.

Spatial bias in cAMP generation determines biological responses to PTH type 1 receptor activation

Alex D. White et al.

Summary: The study showed that similar amounts of cAMP generated by PTHR for similar lengths of time in different cellular locations, plasma membrane and endosomes, mediate distinct physiological responses. This suggests that subcellular signaling location plays a crucial role in achieving specificity in PTHR-mediated biological outcomes and may lead to rational drug design based on spatiotemporal manipulation of GPCR signaling.

Receptor-Arrestin Interactions: The GPCR Perspective

Mohammad Seyedabadi et al.

Summary: Arrestins are a small family of proteins found in most vertebrates, binding to a variety of G protein-coupled receptors and playing key roles in preventing further receptor-G protein coupling, promoting receptor internalization, and initiating specific signaling pathways. Research on the molecular mechanisms of arrestin-GPCR interactions has focused on arrestin elements in receptor binding, but further investigations are needed on the receptor elements involved in arrestin binding to fill existing knowledge gaps. Understanding how receptor elements activate arrestin and contribute to high-affinity binding is crucial for developing arrestin mutants with desired functional characteristics.
