Identification of stomatal-regulating molecules from de novo arylamine collection through aromatic C-H amination

Yosuke Toda et al.

Summary: Stomata are small pores on plant leaves that control gas exchange with the atmosphere. Understanding the mechanism behind stomatal regulation is important for studying plant photosynthesis and drought tolerance. This study used a chemical genetic approach to identify synthetic molecules that influence stomatal movement and discovered a sulfonimidated oxazole derivative (SIM*) that inhibits stomatal opening. The discovery of SIM molecules may lead to the development of agrochemicals for drought tolerance by controlling stomatal movement.

The isothiocyanate sulforaphane induces respiratory burst oxidase homologue D-dependent reactive oxygen species production and regulates expression of stress response genes

Andres Arruebarrena Di Palma et al.

Summary: Sulforaphane (SFN), a phytomolecule found in crucifers, exhibits anticancer properties in animals. This study investigated the signaling pathway of SFN in Arabidopsis and found that SFN induces the production of reactive oxygen species (ROS) through calcium-dependent activation of NADPH oxidase RBOH isoform D. RNA sequencing analysis revealed that SFN upregulates genes associated with heat shock proteins, oxidative stress response, and biotic and abiotic stress response in Arabidopsis.

Identification of Abscisic Acid-Dependent Phosphorylated Basic Helix-Loop-Helix Transcription Factors in Guard Cells of Vicia faba by Mass Spectrometry

Yuki Hayashi et al.

Summary: In guard-cell protoplasts (GCPs) from Vicia faba, an unknown 61 kDa protein is phosphorylated by ABA-activated protein kinase, which binds to 14-3-3 protein in a phosphorylation-dependent manner. ABA-dependent phosphorylated proteins known as AKSs were identified in Arabidopsis thaliana GCPs, but the identity of the 61 kDa protein in Vicia GCPs as an AKS was uncertain. Mass spectrometry analysis revealed several AKS isoforms (VfAKSs) in V. faba, with the 61 kDa protein being identified as VfAKS1, showing the importance of phosphorylation in response to ABA. Co-immunoprecipitation studies also identified interactions between 14-3-3 protein and orthologs of AtABCG40 and CHC1 in guard cells.

Arabidopsis leaf hydraulic conductance is regulated by xylem sap pH, controlled, in turn, by a P-type H+-ATPase of vascular bundle sheath cells

Yael Grunwald et al.

Summary: The study identified the crucial role of AHA2 protein in regulating xylem sap pH and leaf radial water fluxes within the leaf vascular bundle sheath cells. Manipulation of AHA2 activity significantly affected the acid-base balance of xylem sap and leaf hydraulic conductance, demonstrating a causal relationship between AHA2 activity and leaf radial hydraulic water conductance.
