Loss of epigenetic information as a cause of mammalian aging

Summary: All living things experience increased entropy, resulting in the loss of genetic and epigenetic information. In yeast, this loss occurs due to the relocation of chromatin-modifying proteins to DNA breaks, causing cells to lose their identity and age. Through the ICE system, it is found that faithful DNA repair accelerates aging at multiple levels, including the erosion of the epigenetic landscape and DNA methylation. However, this aging process can be reversed by OSK-mediated rejuvenation, supporting the information theory of aging.

Mice embryos lacking Bend3 die at the pregastrulation stage, while Bend3 null embryonic stem cells exhibit severe defects in differentiation, leading to premature activation of hundreds of CpG island-containing bivalent genes. BEND3 is required for maintaining high levels of H3K27me3 at these bivalent genes in order to prevent their premature activation in upcoming developmental stages.

Loss of Slc38a4 imprinting is a major cause of mouse placenta hyperplasia in somatic cell nuclear transferred embryos at late gestation

Summary: Placenta hyperplasia is a common issue in cloned animals and negatively affects the development of cloned embryos. The underlying mechanism behind this phenomenon is largely unknown. In this study, the authors found that placenta hyperplasia occurs in both middle and late gestation in cloned mouse embryos. They further discovered that restoring the expression of a specific amino acid transporter, Slc38a4, can rescue the overgrowth of cloned placentae in late gestation. Molecular analyses revealed that loss of Slc38a4 imprinting leads to over-activation of the mTORC1 signaling pathway, likely due to increased amino acid transport. The findings suggest that loss of Slc38a4 imprinting is responsible for the overgrowth of cloned placentae, and the underlying mechanism involves increased amino acid transport and over-activation of mTORC1.
