Loop extrusion mediates physiological Igh locus contraction for RAG scanning

Hai-Qiang Dai et al.

Summary: RAG endonuclease initiates gene rearrangement in B cells by binding to specific DNA sequences and this process is influenced by chromatin structure. In progenitor B cells, locus contraction promotes gene rearrangement and loop extrusion also drives this process.

High-resolution single-cell 3D-models of chromatin ensembles during Drosophila embryogenesis

Qiu Sun et al.

Summary: This study presents a computational method for reconstructing a large 3D-ensemble of single-cell chromatin conformations from population Hi-C data, applied to study embryogenesis in Drosophila. The modeling of chromatin conformations quantifies chromatin heterogeneity and reveals significant changes during embryogenesis. The findings highlight the importance of high-resolution 3D-ensembles of single-cell chromatin conformations for interpreting population Hi-C data and understanding genome organization.
