Gene-by-gene screen of the unknown proteins encoded on Plasmodium falciparum chromosome 3

The nucleus was found to be the most common location for parasite-specific biology, and four proteins were identified to play essential roles in DNA replication and mitosis. This research demonstrates the feasibility of gene-by-gene screens to uncover the biology of malaria parasites and identify potential drug targets.

The kinetochores in Apicomplexa showed significant divergence in sequence and composition compared to animal and fungal models. The nanoscale organization of kinetochores in Apicomplexa displayed different compartments with distinct biochemical interactions and evolutionary rates. The study also revealed the alignment of kinetochores at the metaphase plate in two different parasites, suggesting a conserved hold signal for preventing premature entry into anaphase. Additionally, the study demonstrated unexpected plasticity in kinetochore composition and segregation between different lifecycle stages of Apicomplexa, indicating diverse requirements to maintain chromosome segregation fidelity.
