SDHB knockout and succinate accumulation are insufficient for tumorigenesis but dual SDHB/NF1 loss yields SDHx-like pheochromocytomas

Neali Armstrong et al.

Summary: Inherited pathogenic succinate dehydrogenase (SDHx) gene mutations cause hereditary pheochromocytoma and paraganglioma tumor syndrome. However, accumulation of succinate and inhibition of 2-oxoglutarate-dependent dioxygenase are insufficient for tumor formation in pheochromocytoma, additional growth-regulatory pathway activation is required.

Succinate as a New Actor in Pluripotency and Early Development?

Damien Detraux et al.

Summary: This article discusses the relationship between different stages of pluripotent cells in embryonic stem cell development and metabolic transitions, and proposes a well-known metabolite, succinate, as a potential player in early pluripotency and embryo development.

A conformational switch in the SCF-D3/MAX2 ubiquitin ligase facilitates strigolactone signalling

Lior Tal et al.

Summary: This study reveals the mechanism of perception and signal activation of the plant hormone strigolactones, as well as the key factors regulating strigolactone signaling. Through the study of mutants, the importance of protein conformational switch in the signaling process is demonstrated. Additionally, an organic acid metabolite that can directly trigger the conformational switch is discovered.

Mass Spectrometry Imaging of Arabidopsis thaliana Leaves at the Single-Cell Level by Infrared Laser Ablation Atmospheric Pressure Photoionization (LAAPPI)

Juha-Pekka Hieta et al.

Summary: This study demonstrates the potential of infrared laser ablation atmospheric pressure photoionization mass spectrometry (LAAPPI-MS) in analyzing the substructures of Arabidopsis thaliana leaves and its application in depth profiling analysis. The results show that LAAPPI-MS can detect variations in the distribution of various metabolites among different substructures of A. thaliana leaves.

Summary: Plant metabolism is more complex than individual microbes, with reactions shifting based on internal and external cues. Research suggests that the design principles of plant metabolism are distinct from microbes, primarily coordinated through transcriptional regulation.

On-tissue boronic acid derivatization for the analysis of vicinal diol metabolites in maize with MALDI-MS imaging

Trevor T. Forsman et al.

Summary: Derivatization reactions with boronic acid improved signal of vicinal diol metabolites in maize tissue for mass spectrometry imaging. High-resolution accurate mass measurements tentatively identified many metabolites, while systematic studies on reaction interference and cross-reactivity were conducted for data interpretation verification.

Two mitochondrial phosphatases, PP2c63 and Sal2, are required for posttranslational regulation of the TCA cycle in Arabidopsis

Youjun Zhang et al.

Summary: This study identified the roles of two mitochondrial phosphatases, Sal2 and PP2c63, along with pyruvate dehydrogenase kinase (PDK), in regulating the tricarboxylic acid (TCA) cycle. PDK specifically regulates the pyruvate dehydrogenase complex (PDC), while PP2c63 nonspecifically regulates PDC. The study also showed that these phosphatases affect flux through the TCA cycle and altered metabolism compromises plant growth in mutant lines.
