Enantioselective NiH-Catalyzed Reductive Hydrofunctionalization of Alkenes

Yuli He et al.

Summary: Enantioselective NiH-catalyzed reductive hydrofunctionalization of olefins has gained much attention in recent years. By using simple chiral ligands, a variety of functionalized olefins and electrophiles can undergo diverse transformations to yield hydrofunctionalized products with high regio- and enantioselectivity. This review discusses the background, major progress and mechanistic investigations of this reaction.

Catalytic Enantioselective α-Alkylation of Amides by Unactivated Alkyl Electrophiles

Xiaoyu Tong et al.

Summary: This study achieved the objective of using a nickel catalyst to perform enantioselective intermolecular alkylations of racemic Reformatsky reagents, resulting in α-alkylated carbonyl compounds. The importance of chiral catalysts in constructing carbonyl compounds with an α stereocenter was further demonstrated. Additionally, detailed mechanistic studies provided insights into key intermediates and elementary steps of the catalytic cycle.

Summary: This study reports a highly chemoselective and enantioselective Csp(3)-Csp(3) XEC reaction catalyzed by flavin-dependent 'ene'-reductases (EREDs). The photoexcitation of the enzyme-templated charge-transfer complex enables the chemoselective reduction of alkyl halides and high enantioselectivity.

Regio- and Enantioselective Ni-Catalyzed Formal Hydroalkylation, Hydrobenzylation, and Hydropropargylation of Acrylamides to α-Tertiary Amides

Lou Shi et al.

Summary: This research presents intermolecular regio- and enantioselective formal hydrofunctionalizations of acrylamides, creating a stereogenic center alpha-position to the newly formed C-sp3-C-sp3 bond for the first time. The use of a newly developed chiral ligand enables electronically-reversed formal hydrofunctionalizations, offering an efficient way to obtain diverse enantioenriched amides with a tertiary alpha-stereogenic carbon center.

Chiral Alkyl Amine Synthesis via Catalytic Enantioselective Hydroalkylation of Enecarbamates

Deyun Qian et al.

Summary: A nickel-catalyzed hydroalkylation method has been developed to assemble a wide range of chiral alkyl amines with high regio- and enantioselectivity from enecarbamates and alkyl halides. The method is suitable for both nonactivated and activated alkyl halides, producing enantiomerically enriched amines with high functional group tolerance.

Enantioselective C(sp3)-C(sp3) cross-coupling of non-activated alkyl electrophiles via nickel hydride catalysis

Srikrishna Bera et al.

Summary: The enantioselective cross-coupling of non-activated alkyl halides with alkenyl boronates enables the synthesis of chiral alkyl boronates, which is a useful method for drug development. This reaction proceeds via nickel hydride insertion into an internal alkene followed by nickel-catalysed alkyl-alkyl cross-coupling.

The Asymmetric Synthesis of Amines via Nickel-Catalyzed Enantioconvergent Substitution Reactions

Ze-Peng Yang et al.

Summary: Chiral dialkyl carbinamines are important compounds in various fields, and two mild methods for their catalytic enantioconvergent synthesis using a chiral nickel catalyst have been reported. These methods are versatile and can be used for the synthesis of a variety of functionalized dialkyl carbinamines, including a one-pot variant for generating enantioenriched compounds from commercially available amino acid derivatives. The utility of these methods has been demonstrated in the efficient catalytic enantioselective synthesis of a range of target molecules.

Enantioselective access to chiral aliphatic amines and alcohols via Ni-catalyzed hydroalkylations

Shan Wang et al.

Summary: Chiral aliphatic amine and alcohol derivatives are difficult to access due to the challenge to differentiate between spatially and electronically similar alkyl groups. By utilizing a nickel-catalyzed enantioselective hydroalkylation, enantioenriched alpha-branched aliphatic acyl amines and esters can be efficiently synthesized with good yields and excellent enantioselectivity.
