So Pathogenic or So What?-A Brief Overview of SIV Pathogenesis with an Emphasis on Cure Research

Adam J. Kleinman et al.

Summary: Curing HIV infection is crucial to improve the quality of life of patients, who currently require lifelong antiretroviral therapy. This review discusses different SIV strains and their animal hosts used in cure research, as well as the life cycle and pathogenesis of HIV/SIV. It also emphasizes the role of chronic immune activation and inflammation in disease progression and comorbidities, comparing pathogenic and nonpathogenic infections in natural hosts of SIVs. Furthermore, various HIV cure strategies, particularly immunological therapies and shock and kill approaches, are explored.

The Youngbloods. Get Together. Hypercoagulation, Complement, and NET Formation in HIV/SIV Pathogenesis

Summary: Chronic T-cell immune activation and inflammation are associated with disease progression in persons with HIV, even in those on antiretroviral therapy. Multiple mechanisms drive HIV-associated immune activation and inflammation, including the virus itself, gut dysfunction, co-infections, proinflammatory lipids, treatment toxicity, comorbidities, and behavioral factors. Targeting new pathways that fuel immune activation and inflammation may improve the clinical course of HIV infection.

Antibody-mediated depletion of viral reservoirs is limited in SIV-infected macaques treated early with antiretroviral therapy

Adrienne E. Swanstrom et al.

Summary: The effectiveness of virus-specific strategies, such as administered HIV-specific mAbs, in targeting cells harboring latent HIV genomes during cART has been limited. Depletion of CD4(+) T cells using anti-CD4 mAb did not significantly delay viral rebound after stopping cART, indicating limited effectiveness in key tissue sites.

The Hitchhiker Guide to CD4+ T-Cell Depletion in Lentiviral Infection. A Critical Review of the Dynamics of the CD4+ T Cells in SIV and HIV Infection

Quentin Le Hingrat et al.

Summary: CD4(+) T-cell depletion is a key feature of AIDS in both HIV and SIV infections, occurring early and being most significant at mucosal sites. The clinical outcome is associated with mucosal CD4(+) T-cell recovery during chronic infection, while immune activation and inflammation play critical roles in CD4(+) T-cell depletion.
