Engineered human hepatocyte organoids enable CRISPR-based target discovery and drug screening for steatosis

Delilah Hendriks et al.

Summary: Organoid models of early liver disease, such as nonalcoholic fatty liver disease (NAFLD), are valuable for target discovery and drug screening. Using human fetal hepatocyte organoids, the first stage of NAFLD, steatosis, was successfully modeled with various triggers. Screening of drug candidates identified compounds that effectively resolved steatosis by repressing de novo lipogenesis. The study also introduced FatTracer, a CRISPR screening platform, to identify steatosis modulators and potential targets.

A framework for fibrolamellar carcinoma research and clinical trials

Timothy A. Dinh et al.

Summary: In this article, the authors discuss the progress made in understanding the mechanisms and treatments of fibrolamellar carcinoma (FLC), a rare hepatic cancer that primarily affects adolescents and young adults. They propose recommendations for future research and clinical trials. Significant advancements in FLC research include a better understanding of oncogenic pathways driven by DNAJ-PKAc, identification of potential therapeutic targets, development of research models, elucidation of immune mechanisms with potential for immunotherapies, and completion of the first multicenter clinical trials for targeted therapy in FLC.

Human liver organoids for disease modeling of fibrolamellar carcinoma

Nicole J. C. Narayan et al.

Summary: In this study, patient-derived organoids were successfully developed and used as a model system to recapitulate the histologic morphology, immunohistochemistry, and transcriptome of fibrolamellar carcinoma (FLC). This model system shows promise for improving our understanding of this rare cancer and for drug testing and development.

DCLK1, a Putative Stem Cell Marker in Human Cholangiocarcinoma

Nevi Lorenzo et al.

Summary: The study evaluated the expression and function of DCLK1 in intrahepatic and perihilar cholangiocarcinoma cells. DCLK1 was identified as a specific CSC marker and its inhibitor showed anti-neoplastic effects. In addition, DCLK1 was found to be highly expressed in tumor tissues and may serve as a serum biomarker for early diagnosis of cholangiocarcinoma.

Deregulation of ribosomal proteins in human cancers

Wendy El Khoury et al.

Summary: The ribosome, composed of rRNAs and RPs, is the site for protein synthesis. RPs have been shown to play roles in various pathological processes, especially tumorigenesis and cell transformation. This review focuses on recent advances in understanding the effects of RPs deregulation in different types of cancer and their roles in regulating tumor cell fate.

Summary: This protocol provides detailed culture conditions for long-term expansion of human fetal hepatocytes as organoids and introduces methods for gene editing using CRISPR-Cas9. These approaches are valuable for disease modeling, gene function studies, as well as research on cellular differentiation and cell division processes.

Fibrolamellar carcinoma: An entity all its own

Allison F. O'Neill et al.

Summary: Fibrolamellar carcinoma is a rare malignant tumor originating from the liver, usually affecting late adolescence and young adulthood. Surgical resection is the mainstay of treatment, but systemic therapies also play a key role in disease control. There are few systemic therapies that have demonstrated proven benefits.

Identification of Novel Therapeutic Targets for Fibrolamellar Carcinoma Using Patient-Derived Xenografts and Direct-from-Patient Screening

Gadi Lalazar et al.

Summary: Therapeutics informed by genomics have not been effective for fibrolamellar carcinoma (FLC). A functional drug screen identified that inhibitors of TOPO1 and HDAC, along with napabucasin, showed efficacy on patient-derived xenografts (PDX) of FLC. Inhibiting Bcl-xL in combination with these drugs also showed synergy in treating FLC.

Summary: This study shows the role of CCK in hepatocellular carcinoma (HCC) and how CCK-BR blockade can reverse the premalignant state of the hepatic extracellular matrix, making it less susceptible to HCC development. Hence, CCK-BR blockade is a novel approach for preventing/treating HCC.
