Construction of thermally robust and porous shrimp ferritin crystalline for molecular encapsulation through intermolecular arginine-arginine attractions

Hai Chen et al.

Summary: A novel method for constructing protein colloid crystals in diluted solution was proposed in this study, utilizing the strength of arginine-arginine interactions to form long-range ordered protein soft materials. The resulting colloid crystals exhibited high thermal stability under high salt and alkaline conditions, with a cavity volume three times larger than that of intrinsic ferritin cavity in each unit cell, suitable for molecular encapsulation.

ATP and large signaling metabolites flux through caspase-activated Pannexin 1 channels

Adishesh K. Narahari et al.

Summary: The study demonstrated that Panx1 channels activated by caspase allow large molecule permeation, including anionic and large-molecule permeants, while also allowing large cationic molecules to permeate at a slower rate. Additionally, Panx1 channels provide a pathway for the flux of ATP and other signaling metabolites, both anionic (glutamate) and cationic (spermidine).

Structure of the full-length human Pannexin1 channel and insights into its role in pyroptosis

Sensen Zhang et al.

Summary: The expression of PANX1 is upregulated by various stimuli in human macrophages that promote pyroptosis, with structural and functional studies revealing its potential role in pyroptosis during immune responses. Cryo-electron microscopy and molecular dynamics simulations support the model of hPANX1 as a homo-heptamer with N-termini and C-termini extending deep into the channel pore funnel, providing insights into the channel gating mechanism.

OPLS4: Improving Force Field Accuracy on Challenging Regimes of Chemical Space

Chao Lu et al.

Summary: The development and validation of the OPLS4 force field, which builds upon previous work with OPLS3e, improved model accuracy on challenging regimes of drug-like chemical space. A novel parametrization strategy for charged species was introduced, leading to improved accuracy on benchmarks assessing small-molecule solvation and protein-ligand binding.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
