Unraveling B cell trajectories at single cell resolution

BepiTBR: T-B reciprocity enhances B cell epitope prediction

Summary: Being able to predict B cell epitopes is crucial for biomedical research and clinical applications. Researchers have observed that candidate B cell epitopes have a higher chance of being immunogenic if they are located near CD4(+) T cell epitopes, known as T-B reciprocity. However, existing prediction algorithms for B cell epitopes have not considered this interesting observation. In this study, the authors developed a linear B cell epitope prediction model, BepiTBR, based on T-B reciprocity. They found that explicitly incorporating the enrichment of predicted CD4(+) T cell epitopes (HLA class II epitopes) in the model significantly improved the prediction of linear B cell epitopes. Interestingly, this positive impact was specific to the enrichment of DQ allele binders. Overall, this work provides valuable mechanistic insights into the generation of B cell epitopes and suggests a new approach to improving B cell epitope prediction in the field.
