Structure-Guided Development of Small-Molecule PRC2 Inhibitors Targeting EZH2-EED Interaction

Daohai Du et al.

Summary: DC-PRC2in-01 destabilizes the PRC2 complex, leading to the degradation of PRC2 core proteins and the decrease of global H3K27me3 levels in cancer cells. It effectively inhibits the proliferation of PRC2-driven lymphomas cells and arrests the cell cycle in the G0/G1 phase.

VprBP directs epigenetic gene silencing through histone H2A phosphorylation in colon cancer

Nikhil Baban Ghate et al.

Summary: This study reveals the role of VprBP in colon cancer, showing that it promotes proliferation of colon cancer cells by phosphorylating H2A and repressing growth regulatory genes. Inhibiting VprBP can impair colon tumor growth by blocking H2A phosphorylation and reactivating a transcriptional program resembling that of normal cells.
