Calpain-2 regulates hypoxia/HIF-induced plasticity toward amoeboid cancer cell migration and metastasis

Veronika te Boekhorst et al.

Summary: This article studies the molecular mechanisms of cancer cell invasion and metastasis under hypoxic conditions and identifies calpain-2 as a key regulator of the transition from collective to amoeboid dissemination in breast and head and neck carcinoma cells. The study reveals that hypoxia induces calpain-2-mediated amoeboid conversion by deactivating beta 1 integrins. Targeted intervention of calpain-2 activity can restore normal cell phenotype and prevent lung metastasis.

Mitochondrial fission links ECM mechanotransduction to metabolic redox homeostasis and metastatic chemotherapy resistance

Patrizia Romani et al.

Summary: Romani et al. found that cells on soft extracellular matrix have increased mitochondrial fission and production of mitochondrial reactive oxygen species, which may contribute to resistance to chemotherapy drugs in breast cancer cells. The mechanical properties of the tissue also influence the response to treatment. The study suggests that a soft extracellular matrix enhances redox homeostasis and resistance to oxidative stress and chemotherapy drugs.

Highly motile cells are metabolically responsive to collagen density

Matthew R. Zanotelli et al.

Summary: The study found that changes in collagen density are correlated with cell migration ability and bioenergetics, with significantly different effects observed in highly motile cells. These results reveal a link between matrix mechanics, migratory phenotype, and bioenergetics.

Mitochondrial Fission and Fusion in Tumor Progression to Metastasis

Dillon P. Boulton et al.

Summary: Mitochondria are highly dynamic organelles that change their shape through fission and fusion processes to adapt to different environments and developmental contexts. Dysregulated fission and fusion promote tumor progression, affecting tumor cell growth, metabolism, motility, and invasion, and impacting metastasis. It has been found that mitochondrial fission positively influences metastatic potential in various types of tumors.

AMPK-dependent phosphorylation of MTFR1L regulates mitochondrial morphology

Lisa Tilokani et al.

Summary: This study identifies MTFR1L as a critical mitochondrial protein that transduces AMPK-dependent metabolic changes through regulation of mitochondrial dynamics. The phosphorylation of MTFR1L by AMPK controls its function in regulating mitochondrial morphology and stress-induced AMPK-dependent mitochondrial fragmentation.

AMPK: An odyssey of a metabolic regulator, a tumor suppressor, and now a contextual oncogene

Vasudevarao Penugurti et al.

Summary: Metabolic reprogramming is a biochemical adaptation that allows solid tumors to tolerate various stresses. Adenosine monophosphate (AMP)-activated protein kinase (AMPK) plays a crucial role in regulating cellular metabolism and energy balance. While AMPK was initially considered a tumor suppressor, recent studies have revealed its oncogenic potential. Therefore, the combination of AMPK inhibitors with anti-metabolite drugs is a promising approach to treat AMPK-driven cancers.

An AMPK phosphoregulated RhoGEF feedback loop tunes cortical flow-driven amoeboid migration in vivo

Benjamin Lin et al.

Summary: In this study, the authors developed cell type-specific imaging and perturbation techniques to investigate the migratory behavior of cells in the fruit fly Drosophila. They found that primordial germ cells (PGCs) use retrograde cortical actin flows for guidance during development. They also identified RhoGEF2 as an important regulator of cortical flow and demonstrated the mechanism by which it is regulated.

The AMPK-MFN2 axis regulates MAM dynamics and autophagy induced by energy stresses

Yongquan Hu et al.

Summary: Energy deprivation triggers mitochondrial fission and autophagy, while also increasing the number of mitochondrial-associated ER membranes (MAMs) through AMP-activated protein kinase (AMPK) activity. AMPK translocates to MAMs and interacts with mitofusin 2 (MFN2) during energy stress, regulating autophagy and MAM dynamics. MFN2 plays a critical role in cell survival and function under energy stress, highlighting the importance of the AMPK-MFN2 pathway in cellular stress response.

Adhesion-mediated mechanosignaling forces mitohormesis

Kevin M. Tharp et al.

Summary: Mitochondria control cell fate by producing energy and signals, and the physical characteristics of the extracellular matrix can also affect mitochondrial function. Cells can sense the physical properties of the extracellular matrix and activate a mitochondrial stress response to adaptively tune mitochondrial function. Adhesion-mediated mechanosignaling may play a role in altered mitochondrial functions observed in aging and cancer.
