Cell cycle-specific phase separation regulated by protein charge blockiness

Hiroya Yamazaki et al.

Summary: Protein phosphorylation regulates the behavior of Ki-67 protein during the cell cycle and its impact on organelle morphology. Mitotic hyperphosphorylation of Ki-67 induces liquid-liquid phase separation and chromosome periphery formation, while reducing phosphorylation suppresses phase separation and leads to nucleolar dissolution.

Siwi cooperates with Par-1 kinase to resolve the autoinhibitory effect of Papi for Siwi-piRISC biogenesis

Hiromi Yamada et al.

Summary: Siwi promotes the phosphorylation of Papi, enhancing its binding ability to Siwi-bound piRNA precursors. Papi mutants lacking the Tudor and auxiliary domains escape coordinated regulation by Siwi and Par-1, while another mutant lacking the auxiliary domain fails to bind piRNA precursors.

Maelstrom functions in the production of Siwi-piRISC capable of regulating transposons in Bombyx germ cells

Yurika Namba et al.

Summary: Mael is necessary for piRISC biogenesis in germ cells, connecting Spn-E and Siwi, and participating in the assembly of nuage. Loss of Mael disrupts the interaction between Spn-E and Siwi, as well as the production of Ago3-piRISC, while Siwi-piRISC is still produced.

DEAD-box polypeptide 43 facilitates piRNA amplification by actively liberating RNA from Ago3-piRISC

Ryo Murakami et al.

Summary: This study identifies DDX43 as a key protein in regulating the piRNA amplification pathway in Bombyx, functionally similar to Vasa. DDX43 contains a KH domain that enhances its ATPase activity, while its helicase core is responsible for Ago3-piRISC interaction and ATP hydrolysis. Both domains are required for maximal DDX43 RNA-binding activity, shedding light on unique collaborations within the pathway.
