Detection of the HIV-1 Accessory Proteins Nef and Vpu by Flow Cytometry Represents a New Tool to Study Their Functional Interplay within a Single Infected CD4+ T Cell

Jeremie Prevost et al.

Summary: HIV-1 Nef and Vpu are important for viral immune evasion, release, and replication. A new method was developed to detect these accessory proteins and their cellular substrates simultaneously. The study found that Vpu cannot compensate for the lack of a functional Nef, which is significant for studying ADCC responses using Nef-defective viruses.

Combined noncanonical NF-κB agonism and targeted BET bromodomain inhibition reverse HIV latency ex vivo

Shane D. Falcinelli et al.

Summary: This study demonstrates the synergy in latent reservoir activation when using inhibitor of apoptosis protein (IAP) antagonists in combination with bromodomain and extraterminal domain protein inhibitors (BETi). It also highlights the importance of HIV transcriptional elongation and splicing as barriers to latent HIV protein expression, providing a rationale for further exploration of IAPi+BETi in animal models of HIV latency.

Inhibitory feedback control of NF-KB signalling in health and disease

Jack A. Prescott et al.

Summary: NF-xB plays critical roles in maintaining cell homeostasis and immune responses, but its deregulation can lead to inflammation and cancer. There are feedback loops in cells that limit and reset NF-xB activation, ensuring precise cellular responses and serving as potential targets for therapeutic intervention.
