Genetic Underpinnings of Carotenogenesis and Light-Induced Transcriptome Remodeling in the Opportunistic Pathogen Mycobacterium kansasii

Summary: In this study, genetic determinants involved in Mycobacterium kansasii (Mk) photochromogenicity were identified. These determinants were found to be related to carotenoid biosynthesis, carotenoid degradation, and monounsaturated fatty acid biosynthesis. Introduction of these genes into non-pigmented Mycobacterium smegmatis resulted in the acquisition of photochromogenicity. Additionally, regulators controlling photochromogenicity and carotenoid breakdown were also identified. Lastly, the study provided insights into the changes in the Mk transcriptome in response to light.

These proteins are thought to play a versatile role as host immune sensors and crucial factors in shaping the outcome of Tuberculosis infection, with the potential to pave the way for new vaccines and therapeutics.

Summary: Current treatment of tuberculosis (TB) typically lasts at least 6 months, with the latent Mycobacterium tuberculosis (Mtb) being tolerant to most anti-TB drugs. A key hypothesis is that drugs targeting the Mtb proteasome (Mtb20S) may shorten treatment time when used in combination with conventional drugs. A series of macrocyclic peptides, including macrocycle 6, have been developed that selectively target the Mtb20S and have the potential to be developed as anti-TB therapeutics.
