Aspergillus fumigatus ffmA Encodes a C2H2-Containing Transcriptional Regulator That Modulates Azole Resistance and Is Required for Normal Growth

Sanjoy Paul et al.

Summary: In this study, we characterized a gene designated ffmA as a sequence-specific transcriptional regulator involved in azole drug resistance and normal growth in Aspergillus fumigatus. Loss of ffmA caused sensitivity to azole drugs and a large reduction in normal growth. Overproduction of the AtrR transcription factor restored growth to ffmA null cells. Our data indicated that FfmA can recognize promoters of genes involved in azole resistance as well as its own promoter. This study sheds light on the mechanisms underlying azole resistance and normal growth in A. fumigatus.

Structural insights into cooperative DNA recognition by the CCAAT-binding complex and its bZIP transcription factor HapX

Eva M. Huber et al.

Summary: The interaction between CBC and HapX controls iron metabolism in fungi. X-ray structural analysis reveals that CBC forms a complex with HapX and DNA, with the interaction between HapX and CBC mediated by DNA. HapX shows structural and functional plasticity, which may also apply to related transcription factors.

Azole Resistance-Associated Regulatory Motifs within the Promoter of cyp51A in Aspergillus fumigatus

Alexander Kuehbacher et al.

Summary: First-line treatment of aspergillosis with azole antifungals is challenged by increasing resistance. This study investigated the regulatory motifs associated with azole resistance in the target gene Cyp51A, revealing the roles of different binding sites under varying iron availability. Understanding these mechanisms is crucial for developing interventions against resistance.

Biochemical Identification of a Nuclear Coactivator Protein Required for AtrR-Dependent Gene Regulation in Aspergillus fumigatus

Sanjoy Paul et al.

Summary: Azole drugs are important for treating Aspergillus fumigatus infections, and resistance to azole drugs is associated with the transcription factor AtrR. This study identified a new protein, NcaA, which is involved in the regulation of AtrR at specific target gene promoters. Understanding the regulation of AtrR function is important for combating azole resistance in this pathogen.

Fungal iron homeostasis with a focus on Aspergillus fumigatus

Matthias Misslinger et al.

Summary: Maintaining iron homeostasis is crucial for fungi to avoid toxic excess and ensure sufficient supply. Iron plays a key role in virulence as pathogens often encounter iron limitation in host niches. Aspergillus fumigatus, a facultative human pathogen, has emerged as a model for studying fungal iron homeostasis.
