Silymarin in cancer therapy: Mechanisms of action, protective roles in chemotherapy-induced toxicity, and nanoformulations

Summary: Silymarin, a milk thistle extract, has various properties, including anti-inflammatory, immunomodulatory, anti-lipid peroxidative, anti-fibrotic, anti-oxidative, and anti-proliferative effects. It not only exhibits anti-cancer functions by targeting multiple molecules involved in cancer hallmarks, but also protects against chemotherapy-induced toxicity. The development of silymarin nanoformulations has addressed its poor water solubility and provides additional benefits such as improved drug loading, sustained release, enhanced cellular uptake, and tumor targeting.

Summary: This study investigated the mechanism underlying the improved absorption of Silymarin (SLM) after oral administration by using nanoparticles and freeze-drying. The results showed that the SLM nanoparticles prepared using high-pressure crystallization method exhibited a narrow size distribution. Furthermore, the freeze-dried SLM nanoparticles with HP-beta-CyD showed improved solubility, wettability, and membrane permeability, leading to enhanced oral absorption of SLM.
