Characterization and Genomic Analysis of a Novel Jumbo Bacteriophage vB_StaM_SA1 Infecting Staphylococcus aureus With Two Lysins

Bingyan Zhang et al.

Summary: The alarming increase in antibiotic resistance in bacterial pathogens has led to a critical need for the development of new antimicrobial agents. A novel phage SA1, isolated from untreated sewage of a pig farm, showed the ability to infect multiple Staphylococcus strains and belonged to the Myoviridae family. The phage SA1 had a jumbo phage genome with 258 ORFs and a serine tRNA. Two proteins, lys210 and lys211, were found to have efficient bactericidal ability. These findings suggest that phage SA1 and lys210/lys211 have the potential to be used as antibiotic agents for treating Staphylococcus infection.

Characterization and Genomic Analysis of PALS2, a Novel Staphylococcus Jumbo Bacteriophage

Yoona Lee et al.

Summary: Staphylococcus aureus is an important human pathogen that can be frequently encountered in clinical and food-processing surroundings. Bacteriophage PALS2, isolated from bird feces, belongs to the Myoviridae family and exhibits extended host inhibition. The genome of PALS2 is classified as a jumbo phage and contains genes involved in DNA replication, nucleotide metabolism, and RNA polymerase, suggesting its potential as a biocontrol agent.

Comparative Genomics of Three Novel Jumbo Bacteriophages Infecting Staphylococcus aureus

Abby M. Korn et al.

Summary: This study introduces three novel S. aureus jumbo phages: MarsHill, Madawaska, and Machias, isolated from swine production environments in the US. These phages belong to a new clade of S. aureus myophages with similar genome size and protein-coding genes, sharing characteristics with known jumbo phages of Bacillus sp. The phages possess genes encoding complete virion and nonvirion RNA polymerases, homing endonucleases, and a retron-like reverse transcriptase, with presumed uracil-substituted DNA hindering DNA sequencing. These phages also have the ability to transduce host plasmids and infect human S. aureus isolates, highlighting their potential impact on genetic diversity and horizontal gene transfer.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

A phage mechanism for selective nicking of dUMP-containing DNA

Tridib Mahata et al.

Summary: Bacteriophages have evolved efficient means to take over the machinery of the bacterial host. Through the study of a bacterial growth inhibitor gene product T5.015 encoded by the T5 phage, it was found that growth inhibition mediated by T5.015 depends on the uracil-excision activity of Ung, leading to DNA replication and cell division arrest.

Isolation and engineering of a Listeria grayi bacteriophage

Stephen Erickson et al.

Summary: The lack of bacteriophages infecting Listeria grayi posed challenges to bacteriophage-based technologies for Listeria. The newly engineered bacteriophage LPJP1 successfully detected low levels of L. grayi with complete specificity and did not infect other bacterial genera. With a genome of over 200,000 base pairs, LPJP1 is the first jumbo bacteriophage reported to infect the Listeria genus.
