Single molecule tracking of bacterial cell surface cytochromes reveals dynamics that impact long-distance electron transport

Grace W. Chong et al.

Summary: The diffusive dynamics of multiheme cytochromes involved in long-distance electron transfer have not been observed or quantified. In this study, the lateral diffusive dynamics of outer membrane-associated multiheme cytochromes MtrC and OmcA in Shewanella oneidensis were investigated using quantum dot labeling, total internal reflection fluorescence microscopy, and single-particle tracking. Confined diffusion behavior was observed along cell surfaces and membrane nanowires, providing evidence for electron transport via overlap of cytochrome trajectories.

Single molecule tracking of bacterial cell surface cytochromes reveals dynamics that impact long-distance electron transport

Grace W. Chong et al.

Summary: In this study, the diffusive dynamics of outer membrane-associated multiheme cytochromes in the metal-reducing bacterium S. oneidensis were observed and quantified for the first time using quantum dot labeling, total internal reflection fluorescence microscopy, and single-particle tracking. The findings revealed that these cytochromes exhibit confined diffusion behavior and can trace a path for electron transport, supporting the long-distance conduction mechanism.

Spatial distribution of biofilm conductivity in a Geobacter enriched anodic biofilm

Abid Hussain et al.

Summary: This study evaluated the spatial distribution of biofilm conductivity (K-bio) on an anodic biofilm at different growth conditions. The increase in K-bio was associated with an increase in current density. Geobacter was the dominant microbial community on all electrode pairs. Acetate concentration significantly influenced the spatial distribution of K-bio.

Long-distance electron transfer in a filamentous Gram-positive bacterium

Yonggang Yang et al.

Summary: Long-distance extracellular electron transfer has been observed in Gram-negative bacteria and now also in a filamentous, unicellular Gram-positive bacterium. The filamentous bacterium Lysinibacillus varians GY32 is capable of bidirectional extracellular electron transfer, forming centimetre-range conductive cellular networks with 1mm-long cells and conductive appendages. Our results demonstrate that Gram-positive bacteria can also participate in long-distance electron transfer processes.

Structure of Geobacter pili reveals secretory rather than nanowire behaviour

Yangqi Gu et al.

Summary: Geobacter species transfer electrons extracellularly through microbial nanowires known as surface appendages. Recent studies have revealed that the nanowires consist of cytochromes OmcS and OmcZ, rather than the previously believed pili composed of PilA-N protein. The use of PilA-C in assembling heterodimeric pili resembles the structure and function of type 2 secretion pseudopili, indicating a potential widespread effect on diverse electron-transferring microorganisms.

Accelerating anaerobic digestion for methane production: Potential role of direct interspecies electron transfer

Zixin Wang et al.

Summary: Anaerobic digestion is a commercial technology used for producing bioenergy from waste, but suffers from inefficiencies. Direct interspecies electron transfer (DIET) is seen as a potential solution to enhance the process, as it allows for faster and more stable transport of reducing equivalents between bacteria and archaea. This review paper discusses different mechanisms of DIET, its role during organic degradation, startup performance, methane production, and potential industrial applications.
