Summary: This study explores a method for studying molecular function in confined environments by packing biomolecules inside virus capsids and monitoring the emission signal of green fluorescent proteins using atomic force microscopy and total internal reflection fluorescence microscopy. The results show that mechanical and electronic quenching can be decoupled by using different tips, revealing insights into the behavior of green fluorescent proteins within virus capsids.
