PIK3CA is recurrently mutated in canine mammary tumors, similarly to in human mammary neoplasia

Maja Louise Arendt et al.

Summary: Biological features of neoplastic disease affecting mammary gland tissue are shared between canines and humans. Research performed in either species has translational value and early phase clinical trials performed in canines with spontaneous disease could be informative for human trials. This study identified PIK3CA as the most commonly mutated gene in canine mammary tumors, with mutations similar to human breast neoplasia. Other mutations in KRAS and PTEN were also identified. This data supports the validity of studying cancer in non-human species.

Ensembl 2022

Fiona Cunningham et al.

Summary: Ensembl is unique in its flexible infrastructure for access to genomic data and annotation. They have focused on expediting annotation of new assemblies via the Ensembl Rapid Release platform, with the greatest annual number of newly annotated genomes released. They also developed a new method for comparative analyses and annotated non-vertebrate eukaryotes for the first time.

Long-read assembly of a Great Dane genome highlights the contribution of GC-rich sequence and mobile elements to canine genomes

Julia Halo et al.

Summary: The study achieved improved genome assembly of a female Great Dane dog using a combination of long- and short-read sequences. The assembly identified new protein-coding genes not present in the reference genome. Additionally, analysis revealed structural variants dominated by retrotransposon insertions/deletions, suggesting ongoing retrotransposon activity as a driver of genetic variation in dogs.

Clinical cancer genomic profiling

Debyani Chakravarty et al.

Summary: Profiling tumors by next-generation sequencing has the potential to improve diagnostic accuracy, assess heritable cancer risk, and guide treatment selection. Through integrative analyses of tumor and germline variants, as well as identifying mutational signatures that influence therapy response, the clinical utility of cancer genomic profiling is being enhanced. Rapid technological innovation and reduced sequencing costs have enabled the profiling of hundreds of cancer-associated genes as part of routine cancer care.

Comparison of the oncogenomic landscape of canine and feline hemangiosarcoma shows novel parallels with human angiosarcoma

Kim Wong et al.

Summary: The study sequenced cancer genes in canine and feline hemangiosarcomas, showing they share genetic similarities with human angiosarcoma, including recurrent mutations in driver genes. Comparison analysis also revealed different mutational profiles in these tumors across tissue sites.

Establishment of Canine Transitional Cell Carcinoma Cell Lines Harboring BRAF V595E Mutation as a Therapeutic Target

Hyojik Jung et al.

Summary: Recent studies have shown that up to 80% of canine urothelial carcinoma patients carry the BRAF V595E mutation, which is similar to the human V600E mutation. Canine TCC cell lines responded differently to BRAF inhibitors, with sorafenib showing greater effectiveness than vemurafenib. These findings provide valuable research materials for developing therapeutic strategies for canine TCC patients.
