The involvement of DDX3X in compression-induced nucleus pulposus pyroptosis

Summary: This study investigated the relationship between DDX3X and nucleus pulposus pyroptosis and found that DDX3X mediates pyroptosis of NP cells by upregulating NLRP3 expression, leading to intervertebral disc degeneration. Summary: Lysophagy, a form of selective autophagy, plays a role in intervertebral disc degeneration (IDD) by inducing lysosomal damage and promoting cell ferroptosis. The G3BP1/TSC2 complex is involved in coordinating lysophagy and dysfunctional complex results in lysosomal damage and cell ferroptosis. Inhibition of mTOR signaling protects against lysosomal damage and cell ferroptosis. These findings provide insights into the relationship between lysophagy and compression-induced cell ferroptosis and suggest potential targets for IDD treatment.

Summary: Investigating the mechanisms of intervertebral disc degeneration (IVD) has revealed a potential treatment for limiting the progression of low back pain. IVD involves widespread inflammation and cell death within the discs, but the exact progression of the condition is unclear. Researchers in China have found that oxidative stress triggers the leakage of mitochondrial DNA (mtDNA) into the cellular fluid, which activates an immune response pathway and leads to the release of inflammatory cytokines. The activation of this pathway is directly linked to the severity of IVD, allowing for potential monitoring of the condition. Administering a drug to block mtDNA leakage in rats has been shown to slow the progression of IVD.

Inflammatory interactions between degenerated intervertebral discs and microglia: Implication of sphingosine-1-phosphate signaling

Summary: The study found that conditioned media from degenerated intervertebral discs activated microglial cells more significantly than those from normal discs, increasing chemotaxis, migration, and release of pro-inflammatory mediators. Additionally, sphingosine-1-phosphate content in the conditioned media from degenerated discs was significantly higher than in normal discs, and FTY720 effectively inhibited the effects of degenerated discs on microglial inflammatory factor transcription and migration.

Targeting miR-10a-5p/IL-6R axis for reducing IL-6-induced cartilage cell ferroptosis
