pH-Activatable cell penetrating peptide dimers for potent delivery of anticancer drug to triple-negative breast cancer

Summary: The designed pH-activatable cell-penetrating peptide dimer LH2 with histidine residues is able to penetrate cells specifically in weak acidic conditions, delivering paclitaxel effectively into triple-negative breast cancer cells. Both non-covalent complexes (PTX-LH2(M)) and covalent conjugates (PTX-LH2(C)) of LH2 show strong antitumor effects in a triple-negative breast cancer grafted mouse model at extremely low dosages, indicating potential for targeted drug delivery and therapy.

The use of thiol-reactive cell-penetrating peptides allows for simple co-incubation to deliver thiol-containing proteins, greatly simplifying protein and antibody delivery with minimal genetic or chemical engineering.

Summary: Biological membrane fusion is a highly specific and coordinated process where vesicular fusion events occur simultaneously. The study demonstrates the specificity of a liposomal fusion model system using lipidated derivatives of de novo designed heterodimeric coiled coil (CC) peptide pairs. Important relationships between fusogenic efficacy and peptide characteristics were revealed, with a new highly fusogenic CC pair (E-3/P1(K)) identified.
