Sucrose intake by rats affected by both intraperitoneal oxytocin administration and time of day

Simone Rehn et al.

Summary: This study examined the role of entrainment on intake patterns, oxytocin's effects on sucrose intakes, and locomotor activity, as well as whether oxytocin's effects were mediated by its actions at the oxytocin receptor. The results suggest that time of day and oxytocin impact sucrose solution intake under daily limited access in rats, and the sedative-like effects of oxytocin should be considered in future studies on oxytocin and food intake.

Vasopressin neurons in the paraventricular hypothalamus promote wakefulness via lateral hypothalamic orexin neurons

Md Tarikul Islam et al.

Summary: PVHAVP neurons in the paraventricular nucleus of the hypothalamus play a role in promoting wakefulness by interacting with orexin/hypocretin neurons in the lateral hypothalamus. These neurons also receive inputs from brain regions involved in sleep-wakefulness regulation, stress response, and energy metabolism, and mediate arousal induced by novelty stress and melanocortin receptor agonist.

The Theta Paradox: 4-8 Hz EEG Oscillations Reflect Both Sleep Pressure and Cognitive Control

Sophia Snipes et al.

Summary: Human electroencephalographic (EEG) oscillations can characterize behavioral and vigilance states. Theta oscillations (4-8 Hz) have been found in opposite conditions of sleep deprivation and alert cognitive control. This study investigated the differences in theta oscillations during cognitive tasks and sleep deprivation. The results suggest that increases in theta oscillations mainly occur in cortical areas not involved in ongoing behavior.

The vagus nerve mediates the suppressing effects of peripherally administered oxytocin on methamphetamine self-administration and seeking in rats

Nicholas A. Everett et al.

Summary: The study found that peripherally administered oxytocin has inhibitory effects on METH self-administration and reinstatement, with a requirement for vagus nerve signaling involvement, and that this vagal dependency is partially mediated by sex and drug withdrawal.

Advances in the field of intranasal oxytocin research: lessons learned and future directions for clinical research

Daniel S. Quintana et al.

Summary: Recent advances in human and animal research provide converging evidence for functionally relevant effects of intranasal oxytocin administration, supporting the idea that direct nose-to-brain delivery underlies the behavioral effects of oxytocin on social cognition and behavior. Furthermore, discussions on previously debated methodological issues, such as pre-registration, reproducibility, statistical power, interpretation of non-significant results, dosage, and sex differences, are integrated with suggestions for the next steps in translating intranasal oxytocin into psychiatric applications.

The influence of oxytocin-based interventions on sleep-wake and sleep-related behaviour and neurobiology: A systematic review of preclinical and clinical studies

Joel S. Raymond et al.

Summary: This study evaluated the influence of oxytocin on sleep-wake outcomes and found that manipulation of the OXT system can affect these outcomes. Preclinical evidence suggests that OXT can promote wakefulness, while clinical evidence shows little to no sleep-promoting effects of OXT. OXT dose was identified as a likely modulatory factor of OXT-induced effects on sleep-wake behavior.

Sleep Quality and Electroencephalogram Delta Power

Siyu Long et al.

Summary: Delta activity on EEG is considered a biomarker of homeostatic sleep drive, with delta power being associated with sleep duration and intensity. This study found that changes in delta power may both decrease and increase for higher sleep quality, influenced by factors such as underlying illnesses, treatment methods, and sleep stages. Patients with certain disorders may exhibit higher or lower delta power, depending on the specific condition, while therapy choices can also impact delta power changes. Additionally, differences in delta power during different sleep stages may also affect sleep quality.

Intranasal oxytocin for Negative Symptoms of Schizophrenia: Systematic Review, Meta-Analysis, and Dose-Response Meta-Analysis of Randomized Controlled Trials

Michel Sabe et al.

Summary: A systematic review and meta-analysis on the effects of intranasal oxytocin for treating negative symptoms in schizophrenia showed inconsistent results, with no consistent beneficial effect found. Higher doses of oxytocin may have a beneficial effect on negative symptoms, but further studies are needed to confirm this. There was no beneficial effect of oxytocin on positive symptoms in the main meta-analysis, although a potential advantage of higher doses was suggested in the dose-response meta-analysis.

Medial Parabrachial Nucleus Is Essential in Controlling Wakefulness in Rats

Qi Xu et al.

Summary: Activation of the medial part of the parabrachial nucleus (MPB) induces continuous wakefulness in rats for 10 hours, while chemogenetic inhibition of PB neurons decreases wakefulness for the same duration. This suggests that glutamatergic MPB neurons play an essential role in controlling wakefulness, and that the MPB-brainfore (BF) and MPB-lateral hypothalamus (LH) pathways are major neuronal circuits involved.

Optogenetic activation of DRN 5-HT neurons induced active wakefulness, not quiet wakefulness

Rika Moriya et al.

Summary: Studies have shown that 5-HT neurons in the DRN play a crucial role in regulating different waking states, with optogenetic activation of these neurons causing rapid transition from non-REM sleep to active wakefulness. Activation of medullary 5-HT neurons, on the other hand, does not seem to affect sleep/wake states or locomotor activity. Further research into the cortical functional connectivity involved in sleep/wake state regulation is encouraged based on these findings.

The multiple faces of the oxytocin and vasopressin systems in the brain

Valery Grinevich et al.

Summary: Traditionally, hypothalamic neuroendocrine cells were classified into magnocellular and parvocellular neurons for hormone release and brain circuit regulation purposes respectively. However, recent studies show these classic projections are outdated, with oxytocin and vasopressin released in the brain from dendrites and distant axon varicosities to modulate information transfer through conventional synapses.
