Multipronged diagnostic and therapeutic strategies for Alzheimer's disease

Madhu Ramesh et al.

Summary: Alzheimer's disease (AD) is a progressive neurodegenerative disorder that contributes significantly to global dementia cases. The pathology of AD is characterized by the accumulation of amyloid beta plaques and tau neurofibrillary tangles. Despite a lack of complete understanding and approved treatments, research has revealed the multifactorial nature of AD. Therefore, developing multipronged approaches targeting multiple biomarkers and targets is crucial for effective diagnosis and therapeutics.

Tau Protein Binding Modes in Alzheimer's Disease for Cationic Luminescent Ligands

Yogesh Todarwal et al.

Summary: The study investigated a ligand developed for Alzheimer's disease-specific detection of amyloid tau using a combination of theoretical methods and experimental spectroscopies. Results showed that the ligand-protein binding occurs at a hydrophobic pocket defined by specific residues, differentiating from the Pick's disease fold.

Thiophene-Based Dual Modulators of A beta and Tau Aggregation

Madhu Ramesh et al.

Summary: This study screened a targeted library of small molecules to modulate Aβ and Tau aggregation, showing effective modulation of Aβ aggregation and inhibition of Tau aggregation by lead thiophene-based molecules. In silico studies helped understand the interaction mechanism of the lead compounds with Aβ and Tau fibrils, while in cellulo studies demonstrated the therapeutic effect of the candidate compound on neuronal cells.
