Structures of oxysterol sensor EBI2/GPR183, a key regulator of the immune response

Summary: Oxysterols can induce the migration of B-lymphocytes and dendritic cells by binding to EBI2 and activating G protein signaling, which is important for adaptive immunity and antibody production. Abnormal EBI2 signaling is associated with inflammatory bowel disease, sclerosis, and infectious disease. The cryo-EM structures of the EBI2-G(i) signaling complex with its agonist 7 alpha,25-OHC and the inactive EBI2 bound to the inverse agonist GSK682753A reveal the binding site of the agonist and a potential ligand entrance site. Mutations in the binding site and G alpha(i) interface impair G protein signaling and cell migration mediated by oxysterols. These findings shed light on the activation mechanism of EBI2 by oxysterol ligands and may guide the development of therapeutic strategies for EBI2-related diseases.

Membrane phosphoinositides regulate GPCR-(3-arrestin complex assembly and dynamics

Summary: This study reveals the role of membrane phosphoinositides (PIPs) in β-arrestin recruitment and GPCR-β-arrestin complex dynamics through cell-based and in vitro biophysical assays. GPCRs are classified into two groups, with one requiring PIP binding for β-arrestin recruitment and the other not. Plasma membrane PIPs enhance the active conformation of β-arrestin and stabilize GPCR-β-arrestin complexes by promoting a fully engaged state. As allosteric modulators of GPCR-β-arrestin complex dynamics, membrane PIPs allow for additional conformational diversity beyond GPCR phosphorylation alone. For GPCRs that require membrane PIP binding for β-arrestin recruitment, this provides a mechanism for rapid β-arrestin release upon GPCR translocation to endosomes, enabling its quick recycling.

Ligand-directed bias of G protein signaling at the dopamine D2 receptor

Summary: G-protein-coupled receptors (GPCRs) are the largest family of drug targets, but the understanding of how drugs act at GPCRs and contribute to the selectivity of G protein recognition is limited. In this study, the authors investigated the G protein selectivity profile of the dopamine D-2 receptor (D-2) and found that D-2 can discriminate between different members of the Gi/o family. The functional selectivity of D-2 varies across its ligands, which engage D-2 with distinct G protein coupling patterns. The study also identified residues in the D-2 binding pocket that shape these ligand-directed biases.
