Summary: In this study, the hepatotoxicity of HFPO-TA and PFOA was compared using a three-dimensional (3D) primary mouse liver spheroid model. The results showed that HFPO-TA exhibited higher bioaccumulation potential, higher LDH leakage, and lower ATP, albumin, and urea secretion compared to PFOA. Transcriptome analysis identified differentially expressed genes involved in cholesterol metabolism, bile acid metabolism, and inflammatory response. Additionally, HFPO-TA increased triglyceride content and decreased total cholesterol content, while inhibiting the expression of genes related to bile acid transport.
