Development of Kilogram-Scale Convergent Liquid-Phase Synthesis of Oligonucleotides

Xuan Zhou et al.

Summary: A novel convergent liquid-phase synthesis has been developed for manufacturing large quantities of oligonucleotide therapeutics, showing promise for treating diseases affecting millions of people. The method allows for the assembly of an 18-mer drug candidate without the need for column chromatography, resulting in similar purity to standard solid-phase synthesis. The process demonstrates potential for eliminating chromatography in the purification steps, enhancing scalability and reducing environmental impact.

A kinase-cGAS cascade to synthesize a therapeutic STING activator

John A. McIntosh et al.

Summary: Introducing molecular complexity in a efficient manner is a challenging goal in modern synthetic chemistry. Artificial biosynthetic pathways utilizing enzymes have the ability to carry out multiple synthetic steps simultaneously, enabling the discovery and production of complex, non-natural molecules. In this study, an enzymatic cascade was discovered and constructed for the synthesis of MK-1454, a potential immuno-oncology therapeutic. By engineering the enzymes involved, multiple steps of the synthesis were successfully carried out in a one-pot reaction.

Catalytic asymmetric and stereodivergent oligonucleotide synthesis

Aaron L. Featherston et al.

Summary: This study introduces a novel method for controlling the stereochemistry of dinucleotide synthesis using chiral phosphoric acid catalysts, achieving control over the stereogenic phosphorous centers and unprecedented levels of diastereodivergence. The method utilizes two different CPA scaffolds to achieve stereodivergence and does not require stoichiometric activators or chiral auxiliaries, making it a convenient and efficient asymmetric catalysis method for the synthesis of diastereomeric dinucleotides and cyclic dinucleotides of interest in immuno-oncology.
