Stabilization and solubilization of difluprednate in aqueous cyclodextrin solution and its characterization for ophthalmic delivery

Summary: This study investigates the degradation kinetics, stability, and solubility of difluprednate in aqueous CD solutions for the development of an aqueous eye drop vehicle for ophthalmic delivery. The results show that difluprednate has maximum stability at pH 5 in aqueous CD solution. HP gamma CD was found to be the best solubilizer and stabilizer among all the CDs tested. The stability was further improved with the combination of HP gamma CD and different polymers.

Effect of pH, temperature and native cyclodextrins on aqueous solubility of baricitinib

Summary: Baricitinib's solubility was found to be pH dependent, with slight solubility at pH 4-12 and better solubility in acidic and strongly alkaline media. The use of cyclodextrins improved the physicochemical properties of baricitinib, and the formation of host-guest complexes was studied. The influence of cyclodextrin structure and baricitinib's ionization state on complex formation and the impact of cyclodextrins on solubility and membrane permeability were analyzed.

Summary: The study showed that local delivery of nanofingolimod combined with NS/PCs transplantation in a mouse model of acute SCI resulted in better improvement in neurological functions and reduced tissue damage. Co-administration of nanofingolimod and NS/PCs not only increased the survival of transplanted cells, but also promoted their fate towards oligodendrocytic phenotype. This approach could be promising for restoring damaged tissues and improving neurological functions in the acute phase of SCI.
