Metabolomics as a tool for the early diagnosis and prognosis of diabetic kidney disease

Pedro R. Pereira et al.

Summary: Diabetic kidney disease is a common complication of diabetes mellitus and a leading cause of end-stage renal disease. Current methods have limitations in detecting non-albuminuric phenotypes of diabetic nephropathy, but metabolomics offers a promising tool for more accurate diagnosis and prediction.

Transition of acute kidney injury to chronic kidney disease: role of metabolic reprogramming

Zijing Zhu et al.

Summary: Acute kidney injury (AKI) is a global public health concern, and the transition from AKI to chronic kidney disease (CKD) is complex and involves multiple mechanisms, with the metabolic reprogramming of renal tubular epithelial cells (TECs) playing a crucial role.

Immunometabolic rewiring of tubular epithelial cells in kidney disease

Sanne van der Rijt et al.

Summary: Kidney tubular epithelial cells play a crucial role in the response to kidney injury and repair, and their metabolic abnormalities may contribute to kidney disease. Receptors of the innate immune system regulate the metabolic processes of these cells, and the interplay between immunometabolism and kidney health should be taken into consideration.

The Warburg Effect Promotes Mitochondrial Injury Regulated by Uncoupling Protein-2 in Septic Acute Kidney Injury

Ran Ji et al.

Summary: The study demonstrates that aerobic glycolysis is enhanced in septic tubular epithelial cells and inhibition of glycolysis can partially restore mitochondrial function. Knockdown of UCP2 results in increased aerobic glycolysis and mitochondrial injury. Targeting aerobic glycolysis may be important in the treatment of septic acute kidney injury.

The dysregulation of metabolic pathways and induction of the pentose phosphate pathway in renal ischaemia-reperfusion injury

Angelique M. L. Scantlebery et al.

Summary: The study identified significant accumulation of cholesterol, phospholipids, and sphingolipids in renal tissue following ischemia-reperfusion injury. Metabolic pathways related to lipid metabolism were found to be downregulated, except for the pentose phosphate pathway which was upregulated 24 hours after injury.

Tubular Cell Glucose Metabolism Shift During Acute and Chronic Injuries

Anna Faivre et al.

Summary: Acute and chronic kidney disease result in significant healthcare costs worldwide. Kidney metabolism undergoes profound changes during injury, potentially affecting fibrosis progression. Glucose metabolism may play a crucial role in both acute and chronic phases of renal disease.

Single-nuclear transcriptomics reveals diversity of proximal tubule cell states in a dynamic response to acute kidney injury

Louisa M. S. Gerhardt et al.

Summary: The study found that even mild to moderate ischemia-reperfusion injury can cause lasting damage, spreading from the cortico-medullary boundary (CMB) to cortical regions, and the remaining failed-repair PTCs may be triggers for chronic disease progression.

Krappel-like factor 6-mediated loss of BCAA catabolism contributes to kidney injury in mice and humans

Sian E. Piret et al.

Summary: The altered cellular metabolism in kidney proximal tubule (PT) cells, particularly involving the transcription factor Krappel-like factor 6 (KLF6), plays a critical role in acute kidney injury (AKI) and kidney fibrosis. Targeting KLF6-mediated suppression of branched-chain amino acid (BCAA) catabolism may serve as a key therapeutic target in AKI and kidney fibrosis, as shown by protective effects in mice and inverse correlations in human chronic kidney disease patients.

Tyrosine and Tryptophan vibrational bands as markers of kidney injury: a renocardiac syndrome induced by renal ischemia and reperfusion study

Gabrielle Nepomuceno et al.

Summary: Renal ischemia and reperfusion-induced changes in molecular markers, such as cysteine/cystine, fatty acids, Collagen, Tyrosine, and Tryptophan, were detected using FT-Raman spectroscopy, showing potential for predicting progression of acute to chronic kidney injury and mortality in cardiovascular diseases. Monitoring Tyrosine and Tryptophan bands at specific wave numbers proved to be a viable and advantageous method for predicting fatality in cardiovascular diseases using the real-time, multiplexing, and minimally invasive advantages of FT-Raman spectroscopy.
