Effects of intracellular calcium accumulation on proteins encoded by the major genes underlying amyotrophic lateral sclerosis

Giovanni De Marco et al.

Summary: This study investigates the relationship between proteins associated with ALS and intracellular calcium accumulation. The findings suggest that intracellular calcium accumulation leads to decreased levels of several proteins and is associated with the activation of protein degradation and autophagy. These findings contribute to understanding the mechanisms of ALS and identifying new therapeutic targets.

Calpain-2 Mediates MBNL2 Degradation and a Developmental RNA Processing Program in Neurodegeneration

Lee-Hsin Wang et al.

Summary: The loss of structure and function of neurons, along with cognitive decline, is commonly observed in neurologic diseases. This study reveals that increased expression of MBNL2 is associated with neuronal maturation and is essential for neuronal morphogenesis and the transition from fetal to adult RNA processing. The findings suggest that calpain-2-mediated MBNL2 degradation and re-induction of developmental RNA processing may contribute to neurodegeneration.

Detecting motor unit abnormalities in amyotrophic lateral sclerosis using high-density surface EMG

Yuichi Nishikawa et al.

Summary: This study detected specific motor unit (MU) abnormalities in people with amyotrophic lateral sclerosis (ALS) using high-density surface electromyography (HD-SEMG). The results showed that ALS patients had abnormal MU firing behavior compared to controls, and these abnormalities were correlated with disease severity.

Enhanced motor cortex output and disinhibition in asymptomatic female mice with C9orf72 genetic expansion

Sona Amalyan et al.

Summary: This study reveals that asymptomatic mice carrying the C9orf72 gene mutation exhibit abnormal motor cortex output. The reduction in the number of pyramidal neurons in the M1 layer 5 and the decreased inhibitory drive in surviving neurons result in higher M1 output, particularly during high-speed animal locomotion. Furthermore, the study shows that the speed-dependent M1 output can predict the likelihood of C9orf72 genetic expansion.

Motoneuron excitability dysfunction in ALS: Pseudo-mystery or authentic conundrum?

Sherif M. Elbasiouny

Summary: Excitability dysfunction of motoneurons in ALS is a disputed topic in the field due to conflicting reports in the literature on its nature, role, and underlying mechanisms. The form and role of excitability dysfunction in the disease are currently unclear, hindering a comprehensive understanding of ALS.
