Chemogenomics identifies acetyl-coenzyme A synthetase as a target for malaria treatment and prevention

Robert L. Summers et al.

Summary: In this study, we identified and validated the Plasmodium falciparum acetyl-coenzyme A synthetase (PfAcAS) as an important, druggable target. Genetic and chemical validation confirmed that mutations in PfAcAS confer resistance to two antiplasmodial compounds and that these compounds directly inhibit the enzyme. We also found that PfAcAS is essential for asexual growth and involved in the epigenetic regulation of gene expression.

Plasmodium falciparum LipB mutants display altered redox and carbon metabolism in asexual stages and cannot complete sporogony in Anopheles mosquitoes

Marco Biddau et al.

Summary: The study highlights the importance of redox regulation in malaria parasites and suggests LipB as a potential target for the development of new transmission drugs. Deletion of lipB in Plasmodium falciparum impacts redox regulators expression, central carbon metabolism, and development in the mosquito, suggesting a role for lipB in these processes. Further research on LA biosynthesis is needed to better understand its role in asexual stages of malaria parasites.

Parallel functional reduction in the mitochondria of apicomplexan parasites

Varsha Mathur et al.

Summary: Gregarines are an early-diverging lineage of apicomplexan parasites that provide insights into the origin and evolution of the group. They exhibit significantly reduced energy metabolism in their mitochondria, but retain distinctive tubular cristae. This parallel reduction in gregarine mitochondria expands the diversity of organisms containing MROs and highlights the role of parallel transitions in apicomplexan evolution.

Plasmodium falciparum Acetyl-CoA Synthetase Is Essential for Parasite Intraerythrocytic Development and Chromatin Modification

Isadora Oliveira Prata et al.

Summary: The unique Acetyl-CoA Synthetase in the malaria parasite Plasmodium falciparum, PfACS, plays a key role in metabolic and epigenetic modifications. Knockdown or inhibition of PfACS impairs parasite growth and reduces cytoadherence. This study highlights PfACS as a potential drug target due to its significant differences from human ACS and central role in plasmodial metabolism.

Dephospho-CoA kinase, a nuclear-encoded apicoplast protein, remains active and essential after Plasmodium falciparum apicoplast disruption

Russell P. Swift et al.

Summary: Malaria parasites have an essential organelle called the apicoplast, where metabolic pathways for various syntheses are housed. Studies show that isoprenoid synthesis appears to be the only essential function of the apicoplast in blood-stage parasites. Additionally, the enzyme DPCK responsible for coenzyme A synthesis is localized to the apicoplast and is essential for parasite survival.
