Biophysical characterization of the interaction of Atg8 with a disordered region of Nup159 involved in selective autophagy of the nuclear pore complex

Summary: The selective degradation of large multiprotein complexes remains elusive, but recent investigations have discovered selective autophagic degradation of the yeast Nuclear Pore Complex (NPC). In this study, the researchers performed biophysical characterization of the interaction between Atg8 and an intrinsically disordered region (IDR) of Nup159, and identified thermodynamic parameters and a negative heat capacity change (Delta C-p degrees) for the interaction. The study demonstrates the potential impact of this coupling in the initial step of NPC-phagy and provides insights into conformational selection processes in the interactions of IDRs with their target proteins.

Vanilloid-dependent TRPV1 opening trajectory from cryoEM ensemble analysis

Summary: The study used thermal titration methods and cryo-EM to elucidate the conformational trajectory of the vanilloid receptor TRPV1 with resiniferatoxin (RTx) bound. The results showed that RTx binding induced a sequential conformational wave in TRPV1, opening the intracellular gate first, followed by selectivity filter dilation and pore loop rearrangement to reach the final open state. Understanding this RTx-mediated long-range allostery in TRPV1 could enhance the therapeutic potential of RTx for pain relief in advanced cancer or knee arthritis.

Heat-dependent opening of TRPV1 in the presence of capsaicin

Summary: The cryo-EM structures of rat TRPV1 at different temperatures reveal the heat-dependent gating mechanism of this sensory receptor. By visualizing the opening of TRPV1 in response to noxious heat in the presence of capsaicin, the study shows that the conformational transitions leading to TRPV1 opening occur stepwise. The research also suggests that a subset of residues forms an interaction network directly involved in heat sensing, providing insights into the molecular principles underlying sensing of physical and chemical stimuli by TRPV1, which may be applicable to other thermal sensing ion channels.
