Discovery of V-0219: A Small-Molecule Positive Allosteric Modulator of the Glucagon-Like Peptide-1 Receptor toward Oral Treatment for Diabesity

Juan M. Decara et al.

Summary: Peptidic agonists of the GLP-1 receptor have been successful in treating type-2 diabetes and obesity by reducing food intake. Small molecules acting as positive allosteric modulators of GLP-1R, such as compound 9, show promise in improving drug efficacy with oral administration and reduced side effects. The identified GLP-1R PAM, particularly compound 9, exhibits significant in vivo activity in reducing food intake and improving glucose handling in rodents, suggesting a potential therapeutic approach for obesity-associated diabetes.

A Small-Molecule Oral Agonist of the Human Glucagon-like Peptide-1 Receptor

David A. Griffith et al.

Summary: Peptide agonists of the glucagon-like peptide-1 receptor (GLP-1R) have been a breakthrough in diabetes therapy, but their use is limited due to the need for injection. In this study, a small-molecule GLP-1R agonist, danuglipron, was discovered that can be taken orally. Danuglipron showed the ability to promote GLP-1R signaling and increase insulin levels in primates, but not in rodents. This discovery opens up the possibility of oral small-molecule therapies targeting the validated GLP-1R for metabolic health.

Exploring new targets and chemical space with affinity selection-mass spectrometry

Renaud Prudent et al.

Summary: Affinity selection-mass spectrometry (AS-MS) is a high-throughput screening technique that enables rapid identification of ligands for specific biomolecular targets, regardless of their binding site or functional effects. It has become a popular approach in drug discovery, particularly in identifying small-molecule ligands for challenging biological targets.

Discovery of an Orally Efficacious Positive Allosteric Modulator of the Glucagon-like Peptide-1 Receptor

Francis S. Willard et al.

Summary: LSN3318839 is a positive allosteric modulator of the glucagon-like peptide-1 receptor (GLP-1R), enhancing the activity of weak metabolite GLP-1(9-36)NH2 and full-length ligand GLP-1(7-36)NH2 through G protein-coupled signaling. Experiments show that the combination of LSN3318839 and GLP-1(9-36)NH2 produces insulin secretion and glucose lowering effects similar to GLP-1(7-36)NH2.

Affinity Mass Spectrometry-Based Fragment Screening Identified a New Negative Allosteric Modulator of the Adenosine A2A Receptor Targeting the Sodium Ion Pocket

Yan Lu et al.

Summary: The use of affinity mass spectrometry (MS) for screening negative allosteric modulators (NAMs) of GPCR targets offers advantages in reducing protein and compound consumption and speeding up data acquisition. The newly identified NAM through this method exhibits a unique binding mode and antagonistic property different from known NAMs, showcasing its potential for guiding the design of novel allosteric modulators targeting class A GPCRs.

Affinity selection-mass spectrometry for the discovery of pharmacologically active compounds from combinatorial libraries and natural products

Ruth N. Muchiri et al.

Summary: Affinity selection-mass spectrometry (AS-MS) was invented to address the high-throughput screening demands of combinatorial chemistry, utilizing binding interactions between ligands and receptors to isolate pharmacologically active compounds from mixtures of small molecules. AS-MS has three main approaches: pulsed ultrafiltration (PUF) AS-MS, size exclusion chromatography (SEC) AS-MS, and magnetic microbead affinity selection screening (MagMASS), with the capacity to screen hundreds of thousands of compounds per day.
