Restoring morphology of light sheet microscopy data based on magnetic resonance histology

Yuqi Tian et al.

Summary: The combination of cellular-resolution whole brain light sheet microscopy (LSM) images with an annotated atlas enables quantitation of cellular features in specific brain regions. A method is presented to combine LSM data with magnetic resonance histology (MRH) to restore the morphology of LSM images to the in-skull geometry. This workflow provides a routine method for streamlined registration of LSM images to MRH, allowing more accurate regional annotations and measurements.

Cross-modal coherent registration of whole mouse brains

Lei Qu et al.

Summary: The study introduces a cross-modal registration method, mBrainAligner, for aligning whole mouse brain images to the standard Allen Common Coordinate Framework atlas. Researchers then built a mouse brain atlas for the fluorescence micro-optical sectioning tomography modality, enabling further research on single-cell mapping and neuron types.

Virtual mouse brain histology from multi-contrast MRI via deep learning

Zifei Liang et al.

Summary: This study demonstrates the use of deep convolutional neural networks to estimate histological staining intensity directly from MRI signals, providing three-dimensional images that closely mimic target histology and enhanced sensitivity and specificity compared to conventional MRI markers. Additionally, the relative contribution of each MRI contrast within the networks can be used to optimize multi-contrast MRI acquisition, serving as a resource for validating novel MRI techniques.

A natural mutator allele shapes mutation spectrum variation in mice

Thomas A. Sasani et al.

Summary: This study used BXD mouse lines to identify loci that influence germline mutagenesis. They found that a quantitative trait locus on chromosome 4 affects the accumulation rate of C>A germline mutations in mice, primarily due to the activity of different alleles of the DNA repair gene Mutyh. These alleles are also present in wild populations, indicating that common genetic variation modulates germline mutagenesis.

A platform for experimental precision medicine: The extended BXD mouse family

David G. Ashbrook et al.

Summary: The expansion of the BXD family of mice to 140 fully isogenic strains has created a uniquely powerful model for precision medicine, with the ability to map heritable traits with high power and precision. The BXD family, which segregates for 6 million common DNA variants, is an effective platform for testing causal modeling and predictive validation in experimental precision medicine.

A multimodal cell census and atlas of the mammalian primary motor cortex

Edward M. Callaway et al.

Summary: This study presents a multimodal cell census and atlas of the mammalian primary motor cortex, integrating various molecular and spatial information to reveal a unified genetic landscape of cortical cell types. The results establish a mechanistic framework of neuronal cell-type organization by linking molecular genetic information with phenotypic properties.

Large-scale electron microscopic volume imaging of interfascicular oligodendrocytes in the mouse corpus callosum

Tatsuhide Tanaka et al.

Summary: Single oligodendrocytes in the central nervous system produce myelin sheaths around multiple axons, and interfascicular oligodendrocytes (IOs) in the corpus callosum of adult mice have morphologically polarized cell bodies with processes of different thicknesses for myelinating axons. The myelin sheaths originating from specific IOs have biased thicknesses and are associated with axonal diameter. Despite potential axonal regulation of myelin sheath thickness, a single axon may also independently influence myelin thickness.

Specific White Matter Tracts and Diffusion Properties Predict Conversion From Mild Cognitive Impairment to Alzheimer's Disease

David B. Stone et al.

Summary: The study found that diffusion measures from seven white matter tracts can predict the conversion to Alzheimer's Disease (AD), with axial diffusivity being the most predictive measure. Additional analyses revealed that specific white matter changes in certain brain regions are the best predictors of conversion from MCI to AD.

Gene-by-environment modulation of lifespan and weight gain in the murine BXD family

Suheeta Roy et al.

Summary: Through experiments on female mice, this study found that a high-fat diet can significantly shorten lifespan. Initial body weight and weight gain are important factors affecting longevity, while the response to a high-fat diet is modulated by gene-environment interactions. Classic serum metabolites may not be strong predictors of longevity.
