Tupaia GBP1 Interacts with STING to Initiate Autophagy and Restrict Herpes Simplex Virus Type 1 Infection

Summary: In this study, it was found that Tupaia GBP1 combines with Tupaia STING, promotes autophagy, and moderately inhibits HSV-1 infection in a IFN-independent manner. Mechanistically, tGBP1 interacted with tSTING, Tupaia sequestosome 1, and Tupaia microtubule associated protein 1 L chain 3 to form a complex that promotes autophagy in response to HSV-1 infection. The antiviral activity of tGBP1 against HSV-1 infection is dependent on the activation of autophagy by tSTING, as shown by the rescue of anti-HSV-1 activity in tSTING knockout cells with overexpression of tSTING or its mutant tSTING-DCTT.
