Surviving death: emerging concepts of RIPK3 and MLKL ubiquitination in the regulation of necroptosis

Summary: Lytic forms of programmed cell death, such as necroptosis, involve cell rupture, release of cellular contents, and triggering of inflammatory responses. Necroptosis has been found to play important roles in human diseases like cancer, infections, and ischaemia/reperfusion injury. Interactions between RIPK1, RIPK3, and MLKL lead to the formation of a death complex called the necrosome, which triggers MLKL-mediated membrane rupture and necroptotic cell death. Post-translational modifications, especially phosphorylation, tightly control necroptotic cell death.

The regulation of gastric cell growth is influenced by key cell cycle regulators, including Cyclins and Cyclin-dependent kinases (CDKs). Dysregulation of these molecules can lead to disturbances in cell cycle dynamics and the development of gastric cancer. Numerous studies have examined the roles of Cyclins and CDKs in gastric cancer, and this review aims to compile evidence of upstream regulators that activate or inhibit Cyclins and CDKs. It serves as a comprehensive resource for future studies and hypothesis generation.

Summary: The study demonstrated that MG treatment inhibited the proliferation of Hep3B, Mahlavu, and HepJ5 cells both in vitro and in vivo. MG increased levels of superoxide and oxidative stress, as well as induced autophagy and lysosome formation. Western blot analysis showed MG activated cleavage of caspase-3 and PARP, modulated levels of Bcl2, Bax, and Bad ligands, and induced apoptosis. MG also induced autophagy with activation of beclin-1, ATG5+12, and conversion of LC3-I to II. Blocking autophagy enhanced MG-induced cytotoxicity in HCC cells, suggesting MG may serve as a therapeutic supplement for human HCC patients.
