Lineage recording in human cerebral organoids

Zhisong He et al.

Summary: A novel dual-channel recording system called iTracer was established for high-resolution lineage tracing, which combines reporter barcodes with inducible CRISPR-Cas9 scarring. The authors applied iTracer to explore clonality and lineage dynamics during cerebral organoid development, identifying a time window of fate restriction and variations in neurogenic dynamics between progenitor neuron families. Additionally, long-term four-dimensional light-sheet microscopy was established to confirm regional clonality in the developing neuroepithelium of cerebral organoids.

Robustness: linking strain design to viable bioprocesses

Lisbeth Olsson et al.

Summary: This review discusses the challenges of robustness in microbial cell factories for sustainable production, especially when subjected to industrial conditions. It describes industrial perturbations, state-of-the-art technologies to counter process variability, and the potential of single-cell studies for improving system robustness. The importance of achieving consistent and comparable quantification of robustness for selecting strains in industrial bioprocesses is highlighted.

GigaAssay - An adaptable high-throughput saturation mutagenesis assay platform

Ronald Benjamin et al.

Summary: The GigaAssay is a modular high-throughput assay system that can measure molecular functions of thousands of genetic variants at once. It has shown promising results in measuring transcriptional activity by using a GFP reporter system. The use of unique molecular identifiers (UMIs) in the GigaAssay has contributed to its high accuracy and positive predictive value.

Robustness: linking strain design to viable bioprocesses

Lisbeth Olsson et al.

Summary: Microbial cell factories are increasingly popular for sustainable chemical production, but face challenges in robustness when scaled up for industrial conditions. This review discusses predictable and stochastic industrial perturbations, advanced technologies to counter process variability, and the potential of single-cell studies to improve system robustness. Consistent and comparable quantification methods for robustness are highlighted for selecting strains in industrial bioprocesses.

The Adaptive Potential of the Middle Domain of Yeast Hsp90

Pamela A. Cote-Hammarlof et al.

Summary: This study systematically quantifies the cost of adaptation along a large stretch of protein sequence by analyzing approximately 2,300 amino acid changing mutations in the middle domain of the heat shock protein Hsp90. The findings show that most beneficial mutations do not have deleterious effects in other environments, except for one specific environment, diamide.

A distinct growth physiology enhances bacterial growth under rapid nutrient fluctuations

Jen Nguyen et al.

Summary: The study reveals that bacterial growth rates can decrease up to 50% when facing nutrient fluctuations with periods as short as 30 seconds to 60 minutes, compared to a stable nutrient environment with equal average concentration. The unique growth response of cells exposed to fluctuating nutrient environments contributes to a 38% reduction in growth rate, suggesting an unexpected adaptation to fluctuations in nutrient availability.
