Mitochondrial dysfunction, oxidative stress, neuroinflammation, and metabolic alterations in the progression of Alzheimer's disease: A meta-analysis of in vivo magnetic resonance spectroscopy studies

Tao Song et al.

Summary: Evidence suggests that metabolic changes in the brain associated with neuroinflammation, oxidative stress, and mitochondrial dysfunction play a key role in the pathophysiology of MCI and AD. Studies showed decreased levels of N-acetyl aspartate and creatine, increased myo-inositol, decreased glutathione, and disrupted energy metabolism. The hippocampus displayed the most significant alterations in these metabolites.

Decreased content of ascorbic acid (vitamin C) in the brain of knockout mouse models of Na+,K+-ATPase-related neurologic disorders

Keiko Ikeda et al.

Summary: The study showed that the alpha 2 and alpha 3 subunits of Na+,K+-ATPase independently contribute to the ascorbic acid levels in the fetal brain, while the alpha 3 subunit plays a role in ascorbic acid transport in the adult basal ganglia and cerebellum. A decrease in ascorbic acid levels may affect neural network development and could be linked to the pathophysiology of ATP1A2- and ATP1A3-related neurologic disorders.

Summary: This study utilized NMR technology to investigate brain metabolic changes in male and female rats induced by prenatal exposure to chlorpyrifos and valproic acid, finding discriminant biomarkers associated with exposure to these compounds and observing that metabolic changes varied with age but not with sex.

Early role for a Na+,K+-ATPase (ATP1A3) in brain development

Richard S. Smith et al.

Summary: Research shows that the ATP1A3 gene is highly expressed in a subset of excitatory neurons during fetal cortical development, and persists in inhibitory neurons, including parvalbumin interneurons, in early postnatal development. The heteromeric Na+,K+-ATPase pump complex can form specific alpha-beta isoform combinations in different cell types, such as alpha 3-beta 1 in excitatory neurons and alpha 3-beta 2 in inhibitory neurons, indicating a cell-type basis for ATP1A3-associated diseases in pre- and postnatal stages of human cortex development.
