Effects of ex vivo blood anticoagulation and preanalytical processing time on the proteome content of platelets

Samuel Tassi Yunga et al.

Summary: This study evaluates the effects of different blood anticoagulants and processing times on platelet function and protein content. The results show that different anticoagulants have time-dependent effects on platelets, and different processing times also result in differences in platelet proteome. These findings are important for hematology studies and mechanistic, translational, and biomarker research.

Dissecting platelet proteomics to understand the pathophysiology of immune thrombocytopenia: studies in mouse models

Patricia Martinez-Botia et al.

Summary: In this study, the researchers investigated the dynamic changes in platelet proteome in ITP mouse models, aiming to simulate the acute and chronic stages of ITP in humans. They found specific protein dynamics associated with alterations in platelet aggregation responses. The study suggests that the platelet proteome could provide diagnostic and prognostic insights into ITP.

The Role of Platelet-Derived Extracellular Vesicles in Immune-Mediated Thrombosis

Alicia S. Eustes et al.

Summary: This review article provides information on the important roles of platelet-derived extracellular vesicles (PEVs) in hemostasis and thrombosis, including their production mechanisms, characteristics, cellular interactions, and pathological role in diseases. The article also highlights the mechanisms through which PEVs can activate parent cells in a feedback loop.

Proteomic and functional profiling of platelet-derived extracellular vesicles released under physiological or tumor-associated conditions

Mauro Vismara et al.

Summary: During hemostasis, thrombosis, and inflammation, activated blood platelets release extracellular vesicles (PEVs) that represent biological mediators of physiological and pathological processes. We have recently demonstrated that the activation of platelets by breast cancer cells is accompanied by a massive release of PEVs, evidence that matches with the observation that breast cancer patients display increased levels of circulating PEVs. Our study compared the PEVs released by platelets upon activation with thrombin and exposure to breast cancer cells, and found that the two populations of PEVs have different protein compositions and elicit different biological responses in target cells.

Increased plasma level of soluble P-selectin in non-hospitalized COVID-19 convalescent donors

Rebecca Mueller et al.

Summary: This study found that the sP-sel plasma concentration remains significantly elevated in COVID-19 convalescent donors weeks after moderate COVID-19, while in vitro platelet function is comparable to non-infected controls. Most COVID-19 convalescent donors showed a gradual decrease in sP-sel levels over a short period.

Platelet removal by single-step centrifugation

Linda G. Rikkert et al.

Summary: A study developed a fast and simple single-step centrifugation protocol for producing high-quality platelet-free plasma for EV research. Compared to the traditional two-step protocol, the single-step protocol is faster, removes platelets effectively, and has a slightly higher plasma yield.

Soluble Platelet Release Factors as Biomarkers for Cardiovascular Disease

Gaukhar Baidildinova et al.

Summary: Platelets play a crucial role in thrombotic diseases, mediating thrombus formation and interacting with vascular cells, inflammatory components, and the coagulation system. Studying markers released by activated platelets in plasma is important for assessing thrombotic risk in different disease conditions.

Protein expression profiling suggests relevance of noncanonical pathways in isolated pulmonary embolism

Vincent Ten Cate et al.

Summary: Patients with isolated pulmonary embolism (PE) have a distinct clinical profile from those with deep vein thrombosis (DVT)-associated PE, with more pulmonary conditions and atherosclerosis. The study identified shared molecular processes between PE phenotypes, as well as an isolated PE-specific protein signature, indicating possible involvement of noncanonical pathways in the acute pathophysiology of isolated PE.

Summary: Extracellular vesicles (EVs) play a vital role in intercellular communication, transferring information between nearby or distant cells. Blood is a major source of EVs, containing various types of EVs. Blood-derived EVs have significant potential in disease diagnosis, treatment monitoring, and prognosis.
