Coupling epigenetics and RNA polyadenylation: missing links

Juncheng Lin et al.

Summary: Precise regulation of gene expression is essential for plant survival. Pre-mRNA polyadenylation, as a cotranscriptional regulatory mechanism, plays a crucial role in fine-tuning gene expression. Epigenetic modification, including DNA methylation, histone modifications, and epitranscriptomic modification, has been found to be involved in regulating polyadenylation in plants. This article summarizes the links between epigenetics and polyadenylation and discusses their novel biological efficacy for plant development and environmental responses.

Reconstitution of 3′ end processing of mammalian pre-mRNA reveals a central role of RBBP6

Moritz Schmidt et al.

Summary: In this study, the endonucleolytic cleavage of eukaryotic mRNA 3' ends followed by polyadenylation was reconstituted using overproduced and purified proteins. A minimal list of 14 essential polypeptides and 2 stimulatory polypeptides for RNA processing were identified. Key findings include the stimulation of cleavage by cleavage factor I and the essential requirement of RBBP6 in contacting and activating the endonuclease CPSF73.

RBBP6 activates the pre-mRNA 3′ end processing machinery in humans

Vytaute Boreikaite et al.

Summary: In this study, the researchers reconstituted the 3' endonuclease activity of human CPSF and identified the protein factors required for this activity. They found that the activation and site-specific pre-mRNA cleavage by CPSF are tightly controlled. The results suggest that fidelity in mRNA processing is maintained through precise regulation of CPSF activity.

Review: Mechanisms underlying alternative polyadenylation in plants - looking in the right places

Arthur G. Hunt

Summary: In recent years, there has been a growing interest in the study of alternative polyadenylation in plants. The connections between the polyadenylation complex and developmental and stress responses are well-established but less is known about the mechanisms that link stimuli with the functioning of the polyadenylation complex. This review focuses on understanding the roles of the polyadenylation complex in plant growth and alternative polyadenylation, as well as exploring other molecular processes that may contribute to regulated polyadenylation.

Wenbin Ye et al.

Summary: movAPA is an R package for modeling and visualizing alternative polyadenylation dynamics across different biological samples, providing functions for preprocessing, annotation, and statistical analysis of poly(A) sites, as well as identification of poly(A) signals, profiling of APA dynamics, and visualization. It offers seven metrics for measuring tissue-specificity or usages of APA sites across samples, and uses three methods for identifying 3' UTR shortening/lengthening events. It also allows exploration of APA site switching involving non-3' UTR polyadenylation.
