Cytidine Triphosphate Synthase Four From Arabidopsis thaliana Attenuates Drought Stress Effects

Moritz Kraemer et al.

Summary: In this study, CTPS4 was found to be upregulated during abiotic stress in Arabidopsis, and CTPS4 knockout mutants showed impaired growth and recovery ability under drought stress. Further investigation revealed the interaction of CTPS4 with other isoforms, affecting cell structure and accumulation of soluble sugars related to drought stress. The study hypothesized that effects on the cell cycle and DNA repair might be relevant for the reduced drought stress tolerance observed in CTPS4 mutants.

Nucleotide imbalance decouples cell growth from cell proliferation

Frances F. Diehl et al.

Summary: Imbalanced nucleotide levels can lead to excessive cell growth, which is not sensed by canonical growth regulatory pathways. Cells rely on replication stress signaling to cope with nucleotide imbalance and maintain cell growth and division.

Mechanisms of feedback inhibition and sequential firing of active sites in plant aspartate transcarbamoylase

Leo Bellin et al.

Summary: ATC in plants is regulated by feedback inhibition via UMP, which binds to a plant-specific loop and blocks the active site, competing with substrate binding. This unique regulatory mechanism of plant ATCs could be utilized to modulate de novo pyrimidine synthesis and plant growth.

Cytosolic CTP Production Limits the Establishment of Photosynthesis in Arabidopsis

Leo Bellin et al.

Summary: CTPS isoforms in the Arabidopsis protein family play crucial roles in early seedling development, particularly CTPS2, which is essential for RNA and DNA synthesis. Knocking down CTPS2 can result in defects in chlorophyll accumulation and photosynthetic performance, and increase sensitivity to DNA-Gyrase inhibitors.

Chloroplast quality control pathways are dependent on plastid DNA synthesis and nucleotides provided by cytidine triphosphate synthase two

Kamran Alamdari et al.

Summary: This study reveals that the ctps2 mutation in Arabidopsis affects chloroplast nucleic acid content by limiting nucleotides for chloroplast genome maintenance, thereby blocking singlet oxygen signaling. Additionally, complementation studies show that CTPS2 is a member of a distinct ancient lineage from CTPS3.

Deciphering CAD: Structure and function of a mega-enzymatic pyrimidine factory in health and disease

Francisco del Cano-Ochoa et al.

Summary: CAD is a 1.5 MDa particle formed by hexameric association of a 250 kDa protein, essential for de novo pyrimidine synthesis pathway in animals. Understanding CAD requires studying the three-dimensional structures, catalytic and regulatory mechanisms of different enzymatic domains, and how they intertwine and function in a coordinated manner.

TOR coordinates nucleotide availability with ribosome biogenesis in plants

Michael Busche et al.

Summary: The TOR protein kinase in plants regulates nucleotide synthesis and metabolic balance by promoting the activity of cytosolic phosphoribosyl pyrophosphate synthetase (PRS4). Knockout of the prs4 gene leads to embryo lethality in Arabidopsis, while silencing of PRS4 expression causes various developmental abnormalities in Nicotiana benthamiana.
