Profiling of the full-length transcriptome in abdominal aortic aneurysm using nanopore-based direct RNA sequencing

Hai Xin et al.

Summary: This feasibility study used the Oxford Nanopore Technologies (ONT) MinION platform to explore the transcriptome profile and molecular mechanisms of abdominal aortic aneurysm (AAA). The study identified novel transcripts and genes, characterized alternative splicing and polyadenylation events, and provided insights into the genome annotation and gene structure of AAA. The study also demonstrated the suitability of ONT methods for identifying long non-coding RNAs (lncRNAs) and revealed differentially expressed transcripts associated with AAA. These findings offer new insights into the pathogenesis of AAA and potential avenues for personalized treatment optimization.

Transcriptome sequencing with nanopore technology for acquiring a deeper understanding of abscisic acid regulation of secondary mechanisms in Salvia miltiorrhiza

Xuejun Kuang et al.

Summary: In this study, the researchers used Oxford Nanopore Technology to investigate the transcriptome variations of Salvia miltiorrhiza in response to ABA treatment. They found that ABA can change gene expression patterns and posttranscriptional splicing to regulate the plant's response to environmental stress. ABA upregulates genes involved in salvianolic acid biosynthesis but has a lesser effect on tanshinone biosynthesis.

Integrative Analysis of Elicitor-Induced Camptothecin Biosynthesis in Camptotheca acuminata Plantlets Through a Combined Omics Approach

Xiang Pu et al.

Summary: Treatments with abiotic elicitors can effectively promote the accumulation of specialized metabolites in plants. This study comprehensively analyzed the elicitation effects of MeJa, AgNO3, and PEG on the biosynthesis of camptothecin (CPT) in Camptotheca acuminata plantlets. The treatments inhibited the photosynthetic pathway and stimulated carbon metabolism and secondary metabolic pathways, resulting in increased CPT levels. The study also discovered new alkaloids, CPT analogs, and precursors, as well as identified key genes involved in CPT biosynthesis.

Identification of WRKY transcription factors involved in regulating the biosynthesis of the anti-cancer drug camptothecin in Ophiorrhiza pumila

Can Wang et al.

Summary: This study demonstrates that OpWRKY6 gene negatively regulates camptothecin biosynthesis in O. pumila. Overexpression of OpWRKY6 significantly reduces camptothecin accumulation, while knockout leads to increased accumulation. Further experiments reveal that OpWRKY6 regulates camptothecin biosynthesis by downregulating the expression of key genes.

Functional characterization of a catalytically promiscuous tryptophan decarboxylase from camptothecin-producing Camptotheca acuminata

Chong Qiao et al.

Summary: The research on the CaTDC3 gene from the Camptotheca acuminata plant reveals that this enzyme exhibits strict stereoselectivity towards L-tryptophan and shows high catalytic activity in decarboxylation. Additionally, CaTDC3 is capable of recognizing different structures of L-tryptophan derivatives and analogs. The study findings demonstrate the significant potential of CaTDC3 in synthetic biology applications.
