Fluorination Increases Hydrophobicity at the Macroscopic Level but not at the Microscopic Level

Weishuai Di et al.

Summary: This study investigates the difference in hydrophobicity between fluorinated hydrophobic polymers at the macroscopic and microscopic levels. It is found that fluorinated polymers exhibit higher hydrophobicity than regular hydrophobic polymers at the macroscopic level, but the hydrophobic effects are equally strong at the microscopic level. This suggests different underlying mechanisms influencing hydrophobicity at different scales.

Fortified Coiled Coils: Enhancing Mechanical Stability with Lactam or Metal Staples

Patricia Lopez-Garcia et al.

Summary: Coiled coils can be stabilized and fine-tuned for thermodynamic and mechanical stability by introducing helix-inducing macrocyclic constraints known as staples. The number of staples affects thermodynamic stabilization against helix uncoiling, while the positioning of staples controls mechanical stability of CCs. Different types, positions, and numbers of staples are key design parameters for using helical macrocyclic templates in emerging biomaterials to adjust CC properties.

Single-molecule observation of mechanical isomerization of spirothiopyran and subsequent Click addition

Ruixiang Yao et al.

Summary: The study found that the ring-opening of STP at room temperature requires forces of around 200-400 pN, and the process of STP transforming into TMC is reversible. Mechanochemically induced intermolecular Click addition was achieved by pulling STP in the presence of maleimide.

Nanomechanics of Lignin-Cellulase Interactions in Aqueous Solutions

Peitao Zheng et al.

Summary: Efficient enzymatic hydrolysis of cellulose in lignocellulose to glucose is crucial for biofuel production, but nonproductive lignin adsorption to cellulase impedes biorefinery development. Investigating the lignin-cellulase interaction, cation-pi interactions were identified and soy protein was shown to be an effective lignin-blocker, improving enzymatic hydrolysis efficiency through electrostatic, cation-pi, and hydrophobic interactions. These findings offer new insights into developing advanced biotechnologies to address nonproductive lignin adsorption, thus enhancing the economics of lignocellulosic biorefinery.

Modulation of a protein-folding landscape revealed by AFM-based force spectroscopy notwithstanding instrumental limitations

Devin T. Edwards et al.

Summary: Single-molecule force spectroscopy was used to study protein folding dynamics, revealing intrinsic changes in protein folding dynamics and transition state height under different pH conditions. Despite differences in destabilization mechanisms, constant-force landscapes showed minimal differences in transition state height, while force-dependent rates increased as pH decreased. This study provides insights for future AFM-based studies of mechanoresponsive proteins.

Effect of Intramolecular Hydrogen Bonds on the Single-Chain Elasticity of Poly(vinyl alcohol): Evidencing the Synergistic Enhancement Effect at the Single-Molecule Level

Yu Bao et al.

Summary: The study investigates the influence of intramolecular hydrogen bonds on the single-chain elasticity of PVA using single-molecule atomic force microscopy, revealing a positive correlation between the average energy of intramolecular hydrogen bonds and the degree of alcoholysis of PVA. Higher degree of alcoholysis results in a stronger synergistic effect on PVA, potentially contributing to its low water solubility at room temperature.

Single molecule force spectroscopy reveals the context dependent folding pathway of the C-terminal fragment of Top7

Jiayu Li et al.

Summary: Top7 is a de novo designed protein with atomic level accuracy that shows a non-cooperative folding process and involves various folding intermediate states. Fragments of Top7 can fold independently in isolation, but their folding pathways are influenced by interactions with neighboring sequences. This context-dependent folding behavior highlights the importance of intra-chain interactions in shaping the folding landscape of Top7, which may have implications for the design of novel proteins.
